【摘要】 目的研究精索静脉曲张对青春期大鼠生精细胞凋亡的影响及生精细胞凋亡与Bcl-2/Bax表达的关系,以探讨精索静脉曲张影响睾丸生精功能的机制。方法通过部分结扎左肾静脉建立青春期大鼠左侧精索静脉曲张模型,采用原位末端脱氧核苷酸转移酶标记法检测睾丸生精细胞凋亡情况及免疫组化SABC法检测Bcl-2/Bax的表达情况。结果试验组细胞凋亡指数显著高于对照组(P<0.05),且双侧睾丸组织内均见大量生精细胞凋亡。试验组Bcl-2表达较对照组低,左侧睾丸比右侧睾丸低;而Bax表达较对照组高,左侧睾丸比右侧睾丸高。结论精索静脉曲张可明显改变青春期大鼠凋亡相关基因Bcl-2/Bax的表达,导致双侧睾丸生精细胞凋亡增加,这可能是精索静脉曲张影响睾丸生精功能的机制之一。更多还原

【Abstract】 Objective To observe the effects of varicocele on the apoptosis of spermatogenic cells in adolescent rats.Methods Experimental varicocele model was established by partial ligation of left renal vein in adolescent male rats.The apoptosis of spermatogenic cells was detected by in situ terminal deoxynucleotityl transferase-mediated dUTP nick end-labeling(TUNEL) technique.The expression of Bcl-2 and Bax was detected by immunohistochemical method.Results The incidence of apoptosis was increased remarkably in experimental group than in control group,and the number of apoptotic spermatogenic cells of bilateral testes was increased.Bcl-2 expression was decreased in experimental group,but Bax expression increased.Conclusion The apoptosis of spermatogenic cells was highly associated with expression of Bcl-2 and Bax in adolescent rats with experimental varicocele,which may lead to male sterility.更多还原
