人血管瘤裸小鼠移植模型形态学动态观察

【摘要】 目的建立人血管瘤裸小鼠异种移植模型并动态观察移植物形态学变化。方法将人婴幼儿血管瘤组织切分为96个小块移植于16只裸小鼠皮下,定期测量瘤体体积,取不同时期移植瘤作HE染色和抗人CD31免疫荧光标记。结果移植瘤大部分(81/96)存活,在移植后1~2个月期间体积明显增大。继而缓慢缩小。组织学观察显示移植瘤早期出现缺血改变,瘤性内皮细胞坏死、间质纤维化,约在移植后1个月出现瘤细胞增生,血管瘤增长,继而瘤内血管扩张,瘤性内皮细胞逐渐减少,血管瘤消退,代以纤维脂肪组织。结论将快速生长的人血管瘤组织移植于裸小鼠体内,移植瘤可存活,其缺血期后生长模式与人血管瘤自然发展过程相似。

【Abstract】 Objective To establish an animal model of human infantile hemangioma on nude mice,and to explore the morphological development of xenografts.Methods The surgically obtained human infantile hemangioma tissue was cut into 96 pieces and then planted into 16 nude mice subcuta- neously,6 grafts each.The size,color,and texture of grafts were observed periodically,and the grafts were harvested at varied intervals for histological examination and anti-human CD31 immunofluores- cent labeling.Results Most of the grafts(81 of 96,84.3%)got engrafted.The size of grafts increased steeply and got to the peak at 2 months,then gradually declined till the end of 6th month.Under light microscopy,the primary tumor was typically that of infantile hemangioma in proliferative phase.During early stage after grafting,degeneration,necrosis,and fibrosis were evident.From about 1 month after grafting,hemangioma tissue grew in area and cellular density.At 45 days,the cellular density got to the peak and karyogenesis was obvious.At 2 months,hemangioma tissue held the most area of grafts,with some of the blood vessels dilatated.In grafts harvested at 3 months later,fibrofatty tissue appeared and got more prominent according to chronological order,with hemangioma tissue involuted.The findings of immunofluorescent labeling proved the chief cellular component of grafts in HE staining sections to be endothelial cells of human origin.Conclusions A xenografted in vivo animal model of human he- mangioma was established on nude mice.The developing pattern of xenografts after survived ischemia resembled a natural course of human infantile hemangioma with rapid growth and subsequent gradual involution.
