无神经节细胞性巨结肠病动物模型的建立

【摘要】 目的用化学方法选择性去除大鼠肠神经节细胞,建立类似于人类无神经节细胞性巨结肠病的大鼠模型。方法通过开腹手术将0.5%的苄基-二甲基-十四烷基氯化铵(BAC)作用于大鼠降结肠远端2 cm长的肠管浆膜表面30 min,用生理盐水作对照组。术后4周处死大鼠,观察作用部位的大体解剖变化,并通过冰冻切片及整装片的免疫荧光和组织化学染色观察局部肠神经节细胞及神经丛的消除情况。结果术后4周的大体解剖发现,在BAC作用处肠管痉挛狭窄,其上方肠管扩张、肠内容物潴留;组织学检查证实,BAC作用处肌间和黏膜下神经节细胞消失,神经纤维明显减少,其乙酰胆碱酯酶的活性明显减低。结论用BAC化学去除法成功地建立了无神经节细胞性巨结肠病的大鼠模型,为进一步研究该病的发病机制和治疗方法提供了适宜的动物模型。

【Abstract】 Objective To set up a rat aganglinonal megaeolon animal model by chemical ablation of enteric plexus.Methods 0.5% benzalkonium chloride(BAC)solution was applied to the serosal surface of the descending colon for 30 minutes to set up this animal model,while saline was applied to the other rats as controls.The rats were sacrificed 4 wk after the procedure for the assessment of mor- phological changes in the BAC-treated and control bowel segments.A detailed histologic examination was performed using fluorescent immunostaining and acetylcholinesterase histochemistry in frozen sec- tions and whole-mount preparations.Results Four weeks after treatment,autopsy revealed a narrow segment at the site of BAC treatment and marked dilation of the proximal segment.By histologic exam- ination,the portion treated with BAC was confirmed to be denervated and decreased acetylcholinester- ase activity;whereas there were normal myenteric and submucous plexuses and acetylcholinesterase activity in the controls.Conclusions Aganglional megacolon can be produced successfully in the rat by applying BAC to ablate enteric plexus.This model may provide the basis for further studies on etiolo- gy,pathophysiology and new treatment of aganglinonal megacolon.
