次级淋巴趋化因子(SLC/CCL21)基因与肿瘤裂解产物构建树突状前列腺癌瘤苗的抗肿瘤免疫作用研究
The antitumor immunity by a dendritic vaccine encoding secondary lymphoid chemokine and tumor lysate on murine prostate cancer
【摘要】 目的探讨转染次级淋巴趋化因子基因的树突细胞(DC)瘤苗在小鼠前列腺癌中的抗肿瘤效应。方法通过脂质体法将次级淋巴趋化因子基因转染至小鼠骨髓来源的树突状细胞,构建 DC 瘤苗;逆转录-聚合酶链反应检测 SLC;种瘤24 d 后 SLC 组的瘤体平均体积为(1430±86)mm~3,45 d 后仍有50%的荷瘤小鼠存活,与其他两组比较差异有统计学意义(P<0.05);免疫组织化学荧光染色可见 SLD 组中 CD4~+、CD8~+T 细胞和 CD11~+DC 的浸润率分别为(15.24±0.67)%、(11.02±0.73)%和(14.50±0.51)%,与 DC 组比较差异有统计学意义(P<0.05)。结论转染SLC 的 DC 瘤苗能有效诱导抗肿瘤的免疫效应,其免疫效应是通过趋化大量 CD4~+、CD8~+T 细胞及CD11~+DC 至肿瘤部位而发挥作用。
【Abstract】 Objective To investigate the antitumor immunity by a dendritic cell(DC)vaccine encoding secondary lymphoid chemokine gene on murine prostate cancer.Methods DCs from bone mar- row of C57BL/6 were transfected with a plasmid vector expressing SLC cDNA by lipofectamine2000 lipo- some.Total RNA extractor from SLC-DC was used to verify the expression of SLC by RT-PCR.The immu- notherapeutic effect of DC vaccine on murine prostate cancer was assessed.Results The SLC plasmid was successfully transfected into DC by verification of RT-PCR.On day 24 after tumor establishment,the aver- age volume of SLC-DC group was 1430 86mm3,on day 45 there was still 50%mice alive,showing a statis- tical difference compared with DC or PBS counterparts(P<0.05).Immunohistochemical staining analysis showed SLD-DC vaccine was(15.24±0.67)%,(11.02±0.73)%and(14.50±0.51)%respective- ly,showing a statistical difference compared with DC counterparts(P<0.05).Conclusion DC vaccine encoding secondary lymphoid chemokine gene can elicit significant antitumor immunity by infiltration of CD4~+,CD8~+ T cell and dendritic cell,which might provide a potential immunotherapy method for prostate cancer.
