表阿霉素预防浅表性膀胱肿瘤术后复发的研究
The Mechanism of intravesical epirubicin instillation against postoperative recurrence of superficial bladder cancer
【摘要】 目的研究表阿霉素预防浅表性膀胱肿瘤术后复发的可能机制。方法通过细胞培养、免疫组织化学研究不同浓度表阿霉素对膀胱肿瘤EJ细胞株的Ki-67的表达的影响,RT-PCR方法检测术前24 h分别接受生理盐水、阿霉素、表阿霉素膀胱灌注患者膀胱肿瘤组织Fas-mRNA的表达。结果0、30、60、90 mg/L表阿霉素各浓度组Ki-67的阳性率分别为79.60±3.50、60.73±3.31、51.57±3.93、45.83±3.75(P<0.01);表阿霉素较生理盐水可以明显促进肿瘤组织中Fas- mRNA的表达(P<0.01)表阿霉素组和阿霉素组比较差异无统计学意义(P<0.05)。结论表阿霉素可能通过下调Ki-67抑制膀胱肿瘤细胞增殖,促进Fas-mRNA的表达诱导凋亡达到抗肿瘤作用。
【Abstract】 Objective To study the possible antitumor mechanism of intravesical epirubicin (EPI) and its efficacy on preventing postoperative recurrence of supedicial bladder cancer.Methods Cell culture and immunohistochemistry were used to assess the effects of different concentrations of epiru- biein (EPI) on the expression of Ki-67 of EJ cell line in vitro,RT-PCR was used to detect the expression of Fas-mRNA of specimens which received intravesical instillatin with normal saline (NS) or adriamycin (ADM) or epirubicin (EPI) respectively 24 h before surgery.Results The positive expression rate of Ki-67 of EJ cells in groups (0,30,60 and 90 mg/L EPI) was 79.60±3.50,60.73±3.31,51.57±3.93 and 45.83±3.75 respectively (P<0.01).The expression of Fas-mRNA in EPI group was significantly increased as compare with NS group (P<0.01),and there was no significant difference between ADM group and EPI group.Conclusion EPI can inhibit the proliferation of tumor cells by down-regulating the expression of Ki-67 and induce apoptosis by promoting the expression of Fas-mRNA.
