1.8GHz微波对紫外线诱发的人淋巴细胞DNA损伤效应的影响

【摘要】 目的研究1.8GHz(比吸收率为3W/kg)微波(MW)对紫外线(UV)诱发的人外周淋巴细胞DNA损伤的影响。方法淋巴细胞采自3名健康青年,体外实验暴露波长为254nm的UV,剂量分别为0.25、0.50、0.75、1.00、1.50和2.00J/m~2;微波暴露0、1.5、4.0h。并设UV与MW的联合暴露组,处理后淋巴细胞再培养0、1.5和4.0h,用彗星试验检测淋巴细胞DNA损伤情况。结果MW组所诱发的DNA损伤与对照组相比,差异无统计学意义(P>0.05);UV所诱发的DNA损伤(平均尾长MTL)分别为(1.71±0.09)、(2.02±0.08)、(2.27±0.17)、(2.27±0.06)、(2.25±0.12)和(2.24±0.11)μm,明显高于空白对照组[(0.96±0.05)μm],差异有统计学意义(P<0.01)。UV与MW的联合暴露组所诱发的MTL,在培养1.5h时,部分剂量组低于相应的UV组;在培养4h时,部分剂量组高于相应的UV组,差异均有统计学意义(P<0.01或P<0.05)。结论1.8GHz微波暴露1.5~4.0h后未见人淋巴细胞DNA损伤增加,但经不同的培养时间,能增强或降低UV所诱发的DNA损伤效应。

【Abstract】 Objective To study whether 1.8 GHz microwaves(MW)(SAR,3 W/kg)exposure can in- fluence DNA damage induced by ultraviolet ray(UV).Methods The lymphoeytes were obtained from three young healthy donors.The cells were exposed to 254 nm UV at the doses of 0.25,0.50,0.75,1.00,1.50 and 2.00 J/m~2.The lymphocytes were also exposed to 1.8 GHz MW(SAR,3 W/kg)for 0,1.5 and 4.0 h.The combi- native exposure of UV plus MW was conducted.The treated cells were incubated for 0,1.5 and 4.0 h.Finally, comet assay was used to detect DNA damage of above treated lymphocytes.Results The difference of DNA damage induced between MW group and control group was not significant(P>0.05).the MTLs induced by UV were(1.71±0.09),(2.02±0.08),(2.27±0.17),(2.27±0.06),(2.25±0.12),(2.24±0.11)μm,respectively,which were significantly higher than that of control[(0.96±0.05)μm](P<0.01).MTLs of some sub-groups in combina- rive exposure groups at 1.5 h incubation were significantly lower than those of corresponding UV sub-groups (P<0.01 or P<0.05).However,MTLs of some sub-groups in combinative exposure groups at 4.0 h incubation were significantly higher than those of corresponding UV sub-groups(P<0.0 1 or P<0.05).Conclusion The ex- posure to 1.8 GHz(SAR,3 W/kg)MW for 1.5 and 4.0 h can not enhance significantly human lymphocyte DNA damage.But MW can reduce or enhance DNA damage of lymphocytes induced by UV at 1.5 h and 4.0 h incu- bation in comet assay in vitro,respectively.
