【摘要】 目的探讨鱼肝油酸钠的脂质体包裹剂型对于人脐静脉内皮细胞(human umbilicalvein endothelial cell,ECV)-304细胞系的作用。方法采用甲基噻唑四唑法(methyl thiazolyltetrazolium,MTT)、透射电镜、DNA 凝胶电泳和流式细胞仪染色等方法观察脂质体鱼肝油酸钠对 ECV-304细胞的作用。结果细胞毒理学显示,脂质体鱼肝油酸钠组活细胞数目下降速度较慢。电镜显示脂质体鱼肝油酸钠组细胞凋亡,凝胶电泳出现典型 DNA 梯度,PI 染色后流式细胞仪检测出现典型亚二倍体峰,凋亡率为22.23%。结论与普通鱼肝油酸钠制剂不同,脂质体鱼肝油酸钠可以促进ECV-304细胞系凋亡。更多还原

【Abstract】 Objective To evaluate the effect of lipo-sodium morrhuate on ECV-304 cell line.Methods The effect lipo-sodium morrhuate was evaluated by toxicology trial(MTF),electron miscroscope,DNA electrophoresis and flow cytometer.Results The toxicology results showed,that the number of vital cells in lipo-sodium morrhuate group decreased slowly.The electron microscope exhibited apoptosis in the lipo-sodium morrhuate group.And there were typical DNA ladder in DNA electrophoresis and typical apoptosis peak in flow cytometer.The apoptosis rate was 22.23%.Conclusions Unlike the normal preparation of sodium morrhuate,lipo-sodium morrhuate could induce apoptosis of ECV-304 cell line.更多还原
