99Tcm-HL91乏氧显像评价小鼠移植瘤放疗疗效
An evaluation on radiotherapy in hepatoma-bearing mice with 99Tcm-HL91 imaging
【摘要】 目的为99Tcm-4,9-二氮-3,3,10,10-四甲基十二烷-2,11-二酮肟(HL91)乏氧显像评价肿瘤放疗疗效提供依据。方法 15只 H22肝癌细胞小鼠移植瘤模型分成3组,每组各5只。A 组不进行放疗;B 组单次放疗25 Gy 后即刻进行99Tcm-HL91显像;C 组单次放疗25 Gy 48 h 后进行99Tcm-HL91显像,计算 B、C 组放疗前后同一感兴趣区(ROI)内肿瘤组织与正常脑组织放射性计数比值(T/N);测定 A、B、C 组凋亡细胞、G1期和 S 期细胞比例及观察肿瘤组织放疗前后病理变化。结果 (1)B 组放疗后 T/N 值较放疗前显著增高(P<0.05);C 组放疗后 T/N 值较放疗前显著降低(P<0.05)。(2)A、B、C 凋亡细胞之间比例差异具有统计学意义(P<0.05,A<B<C)。(3)C 组 S 期细胞所占比例明显低于 A、B 组,G1期细胞比例明显高于 A、B 组(P<0.05);C 组肿瘤组织光学显微镜下可见凋亡细胞和大量细胞碎片。结论 99Tcm-HL91显像是一种有价值的评价放疗疗效的手段。
【Abstract】 Objective One of the factors influencing outcome of radiotherapy was the hypoxic state of the targeted tissue.The possibility of evaluating therapeutic effect during radiotherapy was explored in this experiment using 99Tcm-4,9-diaza-3,3,10,10-tetramethyldodecan-2,11-dione dioxime(HL91)imaging. Methods Fifteen tumor(H22 liver carcinoma)-bearing mice were divided into 3 groups.Group A was a control without radiotherapy.In group B,99Tcm-HL91 imaging was performed immediately after a radiation of 25 Gy.In group C,imaging was performed at 48 h after the radiotherapy.The uptake ratio of tumor to brain(T/N)was calculated before and after radiotherapy in groups B and C.The proportion of apoptosis cells and the cell cycle were assessed with flow cytometry.The pathological changes of tumors were observed under microscope before and after radiotherapy.Results(1)The T/N ratio after radiotherapy was signifi- cantly higher than pre-treatment value in group B(P<0.05).But the T/N ratio dropped below the pre- treatment value at 48 h after radiotherapy(group C,P<0.05).(2)The fraction of apoptosis cells in groups A,B and C were significantly different(P<0.05,A<B<C).(3)The fraction of phase G1 cells in group C was significantly higher,but phase S cells lower,than that in groups A and B.Under microscope,apopto- sis and cell fragments were found in tumor tissue after radiotherapy in group C.However,there was no in- flammatory cell infiltration.Conclusion 99Tcm-HL91 imaging might be valuable in evaluating tumor radio- therapy effects.
