Bmi-1在胃癌中的表达及其意义

【摘要】 目的探讨Bmi-1在胃癌和癌旁组织中的表达水平及其与临床病理特征的关系,初步评价Bmi-1在胃癌发生、发展中的作用及其意义。方法收集20例胃癌患者的癌组织和癌旁组织,采用Western印迹法和RT-PCR检测Bmi-1表达。同时收集2002—2004年146例有3年以上完整随访资料的胃癌术后患者,采用免疫组化法对手术标本进行染色,检测Bmi-1蛋白表达。结果Western印迹和RT-PCR结果均显示,胃癌组织中Bmi-1表达水平明显高于癌旁组织。免疫组化分析也表明,Bmi-1蛋白在胃癌中阳性表达率为67.8%(99/146)。Bmi-1的表达与胃癌大小、临床分期、淋巴结转移和浸润深度密切相关(P<0.05)。而与患者性别、年龄、肿瘤分化程度等无关(P>0.05)。结论Bmi-1在胃痛组织的表达状态与胃癌生长和浸润转移关系密切,可作为反映胃癌生物学行为的有效指标。

【Abstract】 Objective To investigate the association between the expression of B cell-specific MLV integration site-1(Bmi-1)in gastric cancer and its clinicopathologic significance.Methods The samples of cancerous tissues and paracancerous tissues were collected from 20 patients with gastric cancer.The expressions of Bmi-1 protein and mRNA were determined by Western blot and RT-PCR,respectively. The surgical samples from 146 patients with gastric cancer,who were followed up for more than 3 years, were tested by immunohistochemistry for expression of Bmi-1 protein.Results The results of Western blot and RT-PCR demonstrsted that the expression of Bmi-1 in cancerous tissues were significantly high- cr than that in paracancerous tissues.The resuts of immunohistochemical study also showed that the ex- pression of Bmi-1 protein were 67.8%(99/146)in cancerous tissues.Expression of Bmi-1 was highly correlated with tumor size,clinical stage,lymph node metastasis and T classification(P<0.05),but not with sex,age,tumor differentiation(P>0.05).Conclusions Expression ot Bmi-1 in patients with gastric cancer is highly correlated with invasion and metastasis.Bmi-1 may serve as a biologic behavior factor in prognoses of gastric cancer.
