【摘要】 目的采用HPLC-ELSD测定五苓散中泽泻醇A24-乙酸酯、泽泻醇B23-乙酸酯的含量。方法色谱柱:大连依利特Hy-persil C18柱(4.6 mm×250 mm,5μm);流动相:乙腈-水(75∶25);检测器:Alltech2000型蒸发光散射检测器;流速为0.8 mL.min-1;柱温:室温;ELSD气体:高纯氮气;流速为2.0 L.min-1;漂移管温度:82℃。结果在五苓散中测定出的泽泻醇A24-乙酸酯、泽泻醇B23-乙酸酯分别在0.330~1.980μg(r=0.999 5),0.624~4.680μg(r=0.999 4)内呈良好的线性关系。平均回收率分别为101.4%,99.6%,RSD分别为1.7%,3.0%(n=6)。结论采用本法测定五苓散中泽泻醇A24-乙酸酯、泽泻醇B23-乙酸酯的含量,方法简便、可靠、重复性好。更多还原

【Abstract】 OBJECTIVE To establish a method for the determination of alisol A24-acetate and alisol B23-acetate in Wulingsan.METHODS A HPLC combined with evaporation light scattering detector(ELSD)was applied.Chromatographic column was Hypersil C18(4.6 mm×250 mm,5 μm).The mobile phase consisted of acetonitrile-water(75∶25)at the flow rate of 0.8 mL·min-1.The gas flow rate of ELSD was 2.0 L·min-1 and the temperature of drift tube was 82 ℃.RESULTS The linear ranges of the two components were 0.330～1.980 μg(r=0.999 5,n=5)and 0.624～4.680 μg(r=0.999 4,n=5) respectively.The average recoveries of the two components in Wulingsan were 101.4%(RSD=1.7 %) and 99.6%(RSD=3.0%)(n=6),respectively.CONCLUSION The method is simple,practical and reliable to be used for the quality control of the Wulingsan.更多还原
