【摘要】 目的研究兔舌内植入顺铂植入剂体内药物分布特征。方法将顺铂植入剂4mg植入兔舌内,于植药后不同时间点取血和组织样,用原子吸收法测定血和组织样的顺铂浓度,求算药动学参数并与静脉给药比较。结果舌体、颈淋巴结、肾、血液顺铂的峰浓度分别是静脉给药的3.24、6.85、0.57和0.05倍;舌体、颈淋巴结维持有效浓度时间均是静脉给药的720倍以上。结论舌体植入顺铂植入剂舌体和颈淋巴结药物浓度高、有效作用时间长、作用强度大;肾内药物浓度低,血药浓度极低,有利于舌癌治疗。进一步的顺铂植入剂实验室和临床试验值得开展。更多还原

【Abstract】 OBJECTIVE To study the distribution of cisplatin in rabbit after the cisplatin-implants were implanted into rabbit tongue. METHODS A dose of 4 mg cisplatin-implant was implanted into every rabbit tongue, The cisplatin concentrations in plasma and tissue were determined by atomic absorption spectrometry. The pharmacokinetic parameters were calculated and compared with the iv group. RESULTS The peak concentrations in the tongues, neck lymph gland, kidney and plasma were 3.24,6.85,0.57 and 0.045 times as those found in the iv group, respectively. The maintain effective concentration time in the tongue and neck lymph gland was 720 times as those in the iv group. CONCLUSION The cisplatin concentrations in the tongues and neck lymph gland are high, the time that cisplatin acts effectively in rabbit tongue is very long. The cisplatin concentration is low in kidney, and little in plasma.This result of the study expresses the possibility of treating tongue carcinoma with cisplatin-implants, and further studies are highly necessary.更多还原
