氯霉素ELISA检测试剂盒的研制及其性能测试

【摘要】 采用包被多克隆抗体直接竞争ELISA法建立了氯霉素残留的酶联免疫分析方法,并构建了氯霉素残留ELISA检测试剂盒。研究结果表明,所选用的抗体对氯霉素亲合力强、特异性高,试剂盒对氯霉素残留检测的线性范围为0.24~250ng/mL,检测限(IC10)0.47ng/mL,样品的加标回收率70%~130%。试剂盒性能测试结果表明,其准确性和重现性较好,保存期至少6个月,可用于虾肉、鸡肉、蜂蜜、鲜奶样品中氯霉素残留的批量快速、廉价检测。

【Abstract】 A direct competitive enzyme-linked immunosorbent assay(ELISA)with coated polyclonal antibodies (PAbs)was developed for the determination of chloramphenicol(CAP)residue in food of animal origin,and the chloramphenicol ELISA test kit was also constructed. Results of the study indicated that PAbs had high affinity and specificity to CAP. The linear rang of the CAP immunoassay was from 0.24 to 250 ng/mL,with the detection limit(IC10)0.47 ng/mL. And the recoveries of CAP from spiked samples ranged from 70% to 130%. The results of performance measurement demonstrated that the ELISA test kit showed good repeatability and accuracy,and its preserving duration could be 6 months at least. The ELISA test kit could be used for the screening analysis of CAP in a batch of samples such as shrimp,chicken,honey and milk within a short time at low cost.
