黑莓色素大孔吸附树脂纯化工艺及其特性研究

【摘要】 对黑莓色素大孔吸附树脂纯化工艺及其特性进行了研究。通过静态和动态试验,从15种大孔吸附树脂中选出HPD-100A,其纯化黑莓色素效果较好。此外还对纯化的黑莓色素的光谱特性及稳定性进行了研究。结果表明:HPD-100A大孔吸附树脂纯化黑莓色素的最佳工艺参数为:上柱液pH2.0~3.0,样液质量浓度0.67mg/mL,吸附流速1.0mL/min;以60%乙醇为解吸剂,洗脱液流速0.7mL/min;黑莓色素在紫外(219nm)和可见(514nm)光范围内分别有一个明显的吸收峰,该色素在pH1.0~5.0的酸性条件下比较稳定,但最大吸收波长处对应的吸光度值随着pH的增大而呈递减趋势。

【Abstract】 In this paper,the optimum technological parameter of purification and the characteristic of the pigment were studied.The HPD-100A resin was selected from 15 kinds of macroporous adsorption resin by static and dynamic experiment,and its spectral characteristic and stability were studied.The result showed that the optimum technological parameters of HPD-100A resin for blackberry pigment purification were:the pH value of sample solution was about 2.0~3.0,the concentration was 0.67 mg/mL,the flow rate of adsorption was 1.0 mL/min,the desorption solvent was 60% ethanol,and the flow rate of elutant was 0.7 mL/min;Blackberry pigment has two obvious absorption peak which were at 219 nm and 514 nm respectively,it is steady while the pH value was between 1.0 to 5.0,but the maximum absorbancy was minified with the pH value ascended.
