【摘要】 目的检测蛋白激酶B(PKB)在卵巢上皮性癌组织中的表达,探讨其作用机制及临床意义。方法应用逆转录-聚合酶链反应技术(RT-PCR)和western印迹方法对2000年1月至2006年12月在中国医科大学附属第一医院、中国医科大学附属盛京医院、辽宁省肿瘤医院行手术治疗,经病理组织学检查确诊的50例卵巢上皮性癌组织,正常卵巢组织15例,卵巢瘤样病变10例,卵巢良性肿瘤10例,交界性肿瘤10例的PKB的表达进行了分析。结果PKBmRNA在卵巢上皮性癌组织中的半定量检测结果为0.669±0.093,与正常卵巢、卵巢瘤样病变及卵巢良性肿瘤相比,差异无统计学意义(P>0.05);western印迹检测见卵巢上皮性癌组织中磷酸化PKB蛋白(0.496±0.033)与正常卵巢(0.109±0.028)、卵巢瘤样病变(0.121±0.035)及卵巢良性肿瘤组织(0.118±0.023),量化后比较差异有统计学意义(P<0.05),且在Ⅲ～Ⅳ期卵巢癌中的表达水平明显高于Ⅰ～Ⅱ期。结论磷酸化PKB蛋白与卵巢上皮性癌的发生、发展、浸润呈正相关,有望成为卵巢上皮性癌生物学行为的重要指标。更多还原

【Abstract】 Objective To explore the expression of protein kinase B(PKB)in ovarian epithelial cancer and discuss its mechanism and clinical significance.Methods The expression level of PKB was evaluated by RT-PCR and western blot in 50 cases of ovarian epithelial cancer.Results The expression level of PKBmRNA in the ovarian epithelial cancer was 0.669±0.093.Compared with normal ovary,ovarian tumor-like condition and benign ovary tumors,the difference was not significant(P>0.05).There was overexpression of p-PKB in the ovarian epithelial cancer in western blot.Compared with normal ovary,ovarian tumor-like condition and benign ovary tumors,the difference was significant(P<0.05).The expression level in stage Ⅲ～Ⅳ ovarian epithelial cancer was significantly higher than that in stageⅠ～Ⅱ(P<0.05).Conclusion p-PKB may play an important role in carcinogenesis,development and invasion of ovarian epithelial cancer.It may become an important index for the biological behavior of ovarian epithelial cancer.更多还原
