腺病毒介导MnSOD基因转染对香烟烟雾提取物致内皮细胞氧化损伤及凋亡的影响
Effect of adenovirus-mediated MnSOD gene transfection on oxidative injury and apoptosis of endothelial cells induced by cigarette smoking extract
【摘要】 目的探讨香烟烟雾提取物(CSE)对内皮细胞氧化损伤及凋亡的影响,腺病毒介导锰超氧化物岐化酶(MnSOD)基因转染内皮细胞后,能否部分抑制CSE致内皮细胞的氧化损伤及凋亡。方法体外培养人脐静脉内皮细胞株(ECV304),质粒转染MnSOD基因后将内皮细胞分为3组:对照组、空质粒组及MnSOD组,同时分别给予0%、5%及10%CSE刺激。收集不同CSE浓度刺激后的培养上清液和细胞爬片,TBA法检测上清丙二醛(MDA)浓度,TUNEL法检测细胞凋亡率。结果(1)CSE刺激对照组内皮细胞,可致MDA浓度增加和细胞凋亡,存在CSE浓度和时间依赖性。(2)与空质粒组比较,CSE刺激后MnSOD组MDA含量和凋亡减少,有统计学意义(P<0.05)。结论(1)CSE能导致内皮细胞氧化损伤及凋亡。(2)腺病毒介导MnSOD基因可以部分抑制香烟烟雾提取物致内皮细胞氧化损伤及凋亡。
【Abstract】 Objective To elucidate the effect of cigarette smoking extract(CSE)on oxidative injury and apoptosis of endothelial cells and the impacts of adenovirus-mediated MnSOD transfection.Methods The endothelial cells(ECV304)cultured in vitro were divided into the control group,blank plasmid group and MnSOD group after transfection.The cells were harvested and supernatants were collected after interfered by different concentrations of CSE for 24 h.TBA assay was employed to measure the concentrations of malondialdehyde(MDA)and TUNEL assay was conducted to measure AI.Results A.CSE induced dose-dependent and time-dependent increments of MDA and apoptosis of the endothelial cells in the control group(P<0.05).B.CSE induced dose-dependent increments of MDA and apoptosis of the endothelial cells were reduced signticantly in MnSOD group(P<0.05).Conclusion CSE can induce oxidative injury and apoptosis of endothelial cells which could be partly inhibited by transfection of adenovirus-mediated MnSOD.
