定量组织速度成像评价阿霉素心脏毒性左心室功能的变化与心肌细胞凋亡相关性的研究
Correlation Between Changes of Left Ventricular Function and Cardiomyocyte Apoptosis in Adriamycin-induced Myocardial Damage Evaluated by Qantitative Tissue Velocity Imaging
【摘要】 目的研究定量组织速度成像(QTVI)评价阿霉素心脏毒性左心室功能的变化与心肌细胞凋亡的相关性。方法42只兔分成4组:B组10只,每周静脉注射阿霉素2mg/kg,注射2周;C组10只,同样方法给予阿霉素4周;D组12只同样方法给予阿霉素8周;A组(对照组)10只,注射同等剂量生理盐水8周。12周后对4组兔心脏进行QTVI参数测定,同时检测凋亡心肌细胞以及Bcl-2、Bax蛋白的表达。结果C组和D组二尖瓣环平均收缩期峰值速度(Vs)、收缩期加速度(a)和舒张期峰值速度(Ve)明显减低(P<0.01)。A组B组未见凋亡心肌细胞,C组D组心肌细胞凋亡指数明显升高(P<0.01)。C组D组Bcl-2蛋白表达明显低于A组(P<0.01),Bax蛋白表达明显高于A组(P<0.01),Bcl-2/Bax明显低于A组(P<0.01)。B组各参数无变化(P>0.05)。结论心肌细胞凋亡可能是阿霉素心脏毒性左心室功能减低的机制之一。
【Abstract】 Objective To study the relationship between changes of left ventricular function and cardiomyocyte apoptosis in adriamycin-induced myocardial damage in rabbits evaluated with qantitative tissue velocity imaging(QTVI).Methods Fourty-two rabbits were stochastically divided into four groups(10、10、10 and 12 rabbits in groups A,B,C and D respectively),and rabbits in groups B,C and D were given 2 mg/kg body wt.of adriamycin intravenously once a week for 2 weeks,4 weeks and 8 weeks.Ten rabbits of the group A received 2 ml/kg body wt.of normal saline once a week for 8 weeks as the control group.After 12 weeks,QTVI parameters were meas-ured.Apoptotic cardiomyocytes and the expression of Bcl-2 and Bax protein were also detected.Results Average Vs、a and Ve were significantly lower in C and D groups than those of the group A(P<0.01).There were no apoptotic cardiomyocytes in groups A and B;however,the index of apoptic cardiomyocytes was increased significantly in groups C and D(P<0.01).The expression of Bcl-2 protein was siginficantly lower in groups C and D than that in the group A(P<0.01),while the expression of Bax protein was considerably higher in groups C and D than that in the control group(P<0.01).Bcl-2/Bax was significuntly lower in groups C and D than that in the group A(P<0.01).The parameters in the group B were closed to those of the control group(P>0.05).Conclusions Cardiomyocyte apoptosis may be an important mechanism of decreasing left ventricular function due to adriamycin-induced myocardial damage.
