青蒿琥酯抑制人喉癌细胞株Hep-2增殖及对MEK/ERK与NF-κB通路的影响

【摘要】 目的观察青蒿琥酯(Art)对人喉癌细胞株Hep-2增殖及MEK/ERK、核转录因子-κB(NF-κB)信号通路的影响。方法培养人喉癌细胞株Hep-2,四甲基偶氮唑蓝(MTT)比色法、克隆形成实验及流式细胞术测定不同浓度Art对细胞增殖的抑制作用;W estern-b lot测定p-ERK1/2、NF-κB(p65)及NF-κB抑制物(I-κBα)的表达。结果MTT实验、克隆形成实验及流式细胞术均显示,Art明显抑制Hep-2细胞增殖,W estern-b lot显示Art明显抑制p-ERK1/2及NF-κB(p65)表达及增强I-κBα表达。结论青蒿琥酯可抑制人喉癌Hep-2细胞株增殖,其途径与下调MEK/ERK、NF-κB信号通路有关。

【Abstract】 Objective To investigate the effects of Artesunate(Art) on the proliferation of human laryngocarcinoma cell Hep-2 and MEK/ERK,NF-κB signaling pathway. Methods The effect of art on proliferation of Hep-2 cell line was observed by MTT assay,clone formation experiment and flow cytometry;p-ERK1/2,NF-κB(p65) and I-κBα expressions were assessed by Western blot. Results The proliferation of Hep-2 cell line was suppressed by Art;the p-ERK and NF-κB(p65) expressions were inhibited and I-κBα expression was increased by different concentrations of Art. Conclusion Artesunate can depress the proliferation of Hep-2 in the dose dependent manner,the mechanism of which may be due to downregulating MEK/ERK,NF-κB signaling pathway.
