高效液相色谱-质谱/质谱联用法同时测定人血浆中西替利嗪及伪麻黄碱浓度
HPLC-MS/MS determination of cetirizine and pseudoephedrine in human plasma
【摘要】 目的:建立同时测定人血浆中西替利嗪和伪麻黄碱浓度的液相色谱串联质谱法,用以测定健康受试者口服西嗪伪麻缓释片后的血药浓度。方法:血浆样品经沉淀蛋白处理后,以甲醇-水-甲酸为流动相梯度洗脱,采用 Lichrospher C18柱分离,通过电喷雾四极杆串联质谱,以正离子方式检测,SRM 转化后进行定量分析。结果:西替利嗪和伪麻黄碱的线性范围分别为1.032~412.8 ng·mL-1和1.024~819.2 ng·mL-1,提取回收率均大于90%,批内、批间精密度均小于9.7%。两药的定量限可达1 ng·mL-1。结论:本方法专属、准确、灵敏,适用于临床生物样本的分析研究。
【Abstract】 Objective:An HPLC-MS/MS method to simuhaneously determine the concentration of cetirizine and pseudoephedrine in human plasma was established.Methods:Proteins in blood sample were precipitated by metha- nol,separated in Lichrospher Cs column with a mobile phase of methanol- water- formic acid (gradient elution). LC-MS/MS detection was made with an electrospray positive ionization and selected reaction monitoring.Results: The calibration curve was linear over the concentration of l.032 -412.8 ng·mL-1 and 1.024 -819.2 ng·mL-1 for cetirizine and pseudoephedrine respectively.The absolute recoveries were all more than 90%.The intra-and in- ter-batch precisions were less than 9.7% and the lower limits of quantification of the two drugs were each of 1.0 ng·mL-1.Conclusions:The assay is proved to be sensitive,accurate and convenient enough to study the pharma- cokinetic of clinical biological samples.
