RP-HPLC法测定正常人血浆中阿司匹林酯酶活性
RP-HPLC determination of the activity of aspirin esterase in human plasma
【摘要】 目的:建立同时测定血浆中阿司匹林和水杨酸浓度的高效液相色谱法,采用水杨酸浓度与阿司匹林和水杨酸浓度之和的比值表示阿司匹林酯酶的活性。方法:色谱条件:采用 Phenomenex C18(4.6 mm×150 mm,5μm)色谱柱,流动相为乙腈-0.072 mmol·L-1磷酸(22:78,v/v),流速1.0 mL·min-1,检测波长228 nm,柱温30℃。测定阿司匹林和水杨酸浓度,计算水杨酸浓度与阿司匹林和水杨酸浓度之和的比值,绘制概率分布直方图。结果:100名健康人血浆阿司匹林酯酶活性呈正态性分布;33名健康女性和67名健康男性血浆中阿司匹林酯酶活性均显示呈正态性分布。结论:本方法简便、准确、快速,适合于体外血浆中阿司匹林酯酶活性测定研究。
【Abstract】 Objective:To develop an HPLC method for the simultaneous determination of aspirin(ASA)and salicyl- ic acid(SA)in human plasma,and use the ratio of concentration for SA and ASA+SA to express the activity of as- pirin esterase.Methods:The concentration(C)of aspirin and salicylic acid in the plasma were determined by RP- HPLC.Frequency distribution histogram were drawn by calculating CSA/CASA+SA·HPLC condition:the separation was performed on a Phenomenex C18column(4.6 mm×150 mm,5μm),using the mobile phase of acetonitrile-0.072 mmol·L-1phosphorie acid(22:78,v/v)with a flow rate at 1.0 mL·min-1,the detection wavelength was set at 228 nm and column temperature was 30℃.Results:The frequence distribution of aspirin esterase activity of 100 subjects shown normal distribution,with normal distribution for 33 female subjects and 67 male subjects.Conclusion:The method is simple,accurate and rapid,suitable for the determination of aspirin and salicyl- ic acid in plasma.The method can be used for assay the activity of aspirin esterase.
