超临界CO2萃取-气相色谱-质谱联用分析茅苍术挥发性成分
GC-MS analyzing the volatile components of Atractylodes lancea (Thunb.)DC.with supercritical CO2 fluid extraction
【摘要】 目的:研究茅苍术挥发性成分的萃取和分析方法,为建立茅苍术道地性的质量评价体系提供新方法和基础性研究依据。方法:采用超临界 CO2萃取(SFE-CO2)与气相色谱-质谱联用技术分析茅苍术药材中挥发性成分,并对 SFE-CO2萃取压力、温度、时间及 SFE-CO2流量等条件进行优化。结果:萃取压力为25 MPa,萃取温度为45℃,CO2流量为3 L·min-1,萃取时间为3.5 h 时萃取效果最佳。测得 SFE-CO2萃取物84个峰,鉴定出67种化合物。结论:本方法所得结果与传统的提取方法(水蒸气蒸馏法)比较,相对含量高的成分基本一致,且 SFE-CO2萃取法具有萃取率高、萃取成分多、温度低、时间短、无溶剂残留等优点,可用于茅苍术挥发性成分的萃取和分析。
【Abstract】 Objective:To study extraction and analytical method of the volatile components of Atractylodes lancea (Thunb.) DC.,for providing a new method and research foundation to establish genuine quality evaluation system of Atractylodes lancea (Thunb.) DC.Methods:The volatile components of Atractylodes lancea (Thunb.) DC.was analyzed with supercritical CO2 fluid extraction (SFE-CO2) followed by gas chromatography and mass spectrometry (GC-MS),and optimized the conditions of SFE-CO2 on extraction pressure,temperature,time and SFE-CO2 flow rate.Results:When extraction pressure was 25 MPa,extraction temperature was 45℃,SFE-CO2 flow rate was 3 L·min-1 and extraction time was 3.5 h,the effect was the best.We detected 84 peaks in all,identified 67 chemical compounds.Conclusion:The result with this method compared with the orthodox extraction method(steam distillation,SD),relative amount high components are almost coincident,and SFE- CO2 have good qualities as high extraction rate,more extraction components,low extraction temperature,short extraction time and without solvent re- sidual and so on,which can be used to extract and analyze the volatile components of Atractylodes lancea (Thunb.) DC
