HPLC-ELSD法测定麦芽糖中葡萄糖和麦芽三糖等有关物质的含量
HPLC-ELSD determination of glucose,maltotriose and other related substances in maltose
【摘要】 目的:建立 HPLC-ELSD 测定麦芽糖中葡萄糖和麦芽三糖等有关物质的方法。方法:采用 SHISEIDO CAPCELL NH2柱(4.6 mm×150 mm,5μm)色谱柱;流动相:乙腈-水(65:35);流速:1.0 mL·min-1;检测器:蒸发光散射检测器检测;柱温:室温。结果:麦芽糖和葡萄糖及麦芽三糖等有关物质分离度良好,葡萄糖和麦芽三糖的最低检出限分别为0.047μg和0.042μg。结论:本法简便、准确,专属性好,灵敏度高,可用于麦芽糖中葡萄糖及麦芽三糖等有关物质的检查。
【Abstract】 Objective:To establish an HPLC-ELSD method to determine glucose,maltotriose and other related substances in maltose.Method:The HPLC method was conducted on a SHISEIDO CAPCELL NH2 column(4.6mm ×150mm,5μm)and coupled with evaporative light-scattering detection(ELSD),with acetonitrile-water(65: 35)as mobile phase.The flow rate was 1.0 mL·min-1and the column temperature was room temperature.Results: The resolution between glucose,maltotriose and maltose was satisfactory,and the LOD of glucose and maltotriose were 0.047μg and 0.042μg,respectively.Conclution:A simple,accurate and sensitive HPLC method to deter- mine glucose,maltotriose and other related substances in maltose is established.
