RP-HPLC法同时测定丹红粉针中丹参素、原儿茶醛、红花黄色素A和丹酚酸B的含量
Simultaneous determination of the contents of Danshensu,Protocatechuic Aldehyde,Saffior Yellow A and Salviamolic acid B in Danhong powder for injection by RP-HPLC
【摘要】 目的:建立 RP-HPLC 法测定丹红粉针中丹参素、原儿茶醛、红花黄色素 A 和丹酚酸 B 等4种有效成分的含量。方法:采用 Apollo-C18(250 mm×4.6 mm,5μm)色谱柱;以1%冰醋酸-乙腈为流动相梯度洗脱;流速1.0 mL·min-1;柱温25℃;检测波长280 nm。结果:丹参素、原儿茶醛、红花黄色素 A 和丹酚酸 B 浓度分别在1.12~11.2,0.256~2.56,3.46~34.6,36.2~362μg·mL-1范围内与峰面积线性关系良好(r≥0.9995),方法平均回收率分别为99.5%、101.0%、98.8%和100.8%(RSD<3.0%,n=9)。结论:本方法简便、准确,重复性好,可用于同时测定丹红注射剂中丹参素、原儿茶醛、红花黄色素 A 和丹酚酸 B 的含量。
【Abstract】 Objective:To develop a RP-HPLC method for the simultaneous determination of 4 water-soluble components,danshensu,protocatechuic aldehyde,safflor yellow A and salviamolic acid B in danhong power for in- jection.Method:The analysis was carried on a Apollo-C18(250mm×4.6mm,5μm)column.The mobile phase was composed of acetonitrile and 1% acetic acid with gradient elution and the flow rate was 1.0 mL·min-1.The detection wavelength was at 280 nm and column temperature was 25℃.Result:The linear ranges of danshensu, protoeateehuic aldehyde,safflor yellow A and salviamolic acid B were 1.12-11.2,0.256-2.56,3.46-34.6 and 36.2-362μg·mL-1(r≥0.9995),respectively.The average recoveries were 99.5%,101.0%,98.8% and 100.8%(RSD<3.0%,n=9),respectively.Conclusion:This method is simple and can be used to determine the above 4 water-soluble components with satisfactory accuracy and reproducibility.
