高效液相色谱法检测人血浆中甲砜霉素及其药动学研究

【摘要】 目的:建立检测人血浆甲砜霉素的高效液相色谱法,并对其在人体内的药代动力学进行研究。方法:以 ZORBAX SB-C18(4.6 mm×250 mm,5μm)为色谱柱;乙腈-水(30:70)为流动相,流速0.8 mL·min-1;以10,11-二氢-10-羟基-卡马西平(MHD)为内标,血浆样品经乙酸乙酯和正己烷提取后,在225 nm 波长下进行检测。结果:甲砜霉素在0.1~10.0 mg·L-1浓度范围内线性关系良好(r=0.9999),定量下限为0.1 mg·L-1;0.25,1.0,5.0μg·mL-13种浓度的日间、日内精密度RSD(n=5)均小于10%;相对回收率均在85%~115%范围内,绝对回收率大于75%。结论:本方法操作简便,结果准确;适用于血浆甲砜霉素的检测。

【Abstract】 Objective:To establish an HPLC method for the determination of thiamphenicol in human plasma and study of its pharmacokinetics in Chinese volunteers.Method:The analytical column was packed with ZORBAX SB -C18(4.6 mm×250 mm,5 μm),the mobile phase was acetonitrile-water(30:70),the flow rate was O.8 mL· min-1,and detection wavelength was set at 225 nm,respectively.Plasma samples were extracted with ethyl acetate and n-hexane.Results:The linear range of thiamphenicol in human plasma was 0.1-10.0 mg·L-1(r= 0.9999).The lowest limit was 0.1 mg·L-1;Intra-day and inter-day RSD for the analysis of thiamphenicol in plasma samples at three concentration levels were both lower than 10%.Relative and absolute recoveries were 85% -115% and more than 75%,respectively.Conclusion:The HPLC method is simple,high selective and sensitive for the determination of thiamphenicol in plasma.
