云南高峰黄牛BoLA-DRB3基因外显子2的PCR-RFLP多态性分析

【摘要】 通过PCR扩增获得云南高峰黄牛BoLA-DRB3基因第2外显子284bp的特异性条带,用核酸限制内切酶HaeⅢ,RsaⅠ,BstYⅠ和TaqⅠ对其进行消化分析。其中HaeⅢ酶切位点存在9种基因型,由A,B,D,E和F 5种复等位基因控制;RsaⅠ酶切位点存在46种基因型,由A,B,C,D,E,F,G,H,I,J,P,L,M,N,U,V,W和Y 18种复等位基因控制;BstYⅠ酶切位点存在10种基因型,由A,B、C,D和E 5种复等位基因控制;TaqⅠ酶切位点只出现了一种基因型。分析发现云南高峰黄牛DRB3基因第2外显子的61,70,94,104,124,150,154,163,173,182,202和217位的碱基表现出了多态性。经x2检验表明,BstYⅠ和TaqⅠ酶切位点已经达到了Hardy-Weinberg平衡状态,而HaeⅢ和RsaⅠ两个酶切位点未达到Hardy-Weinberg平衡状态。

【Abstract】 In this paper,the exon 2 of BoLA-DRB3 gene was amplified and a uniform fragment of 284bp was obtained in Yunnan Humped Cattle.The 284bp PCR product was digested with restriction endonuclease HaeⅢ,RsaI,BstYI and TaqI,and genetic polymorphism was investigated with PCR-RFLP.9 kinds of genotypes and 5(A,B,D,E,F) alleles were found with enzyme HaeⅢ;46 kinds of genotypes and 18(A,B,C,D,E,F,G,H,I,J,P,L,M,N,U,V,W,Y) alleles were found with enzyme RsaⅠ;10 kinds of genotypes and 5(A,B,C,D,E) alleles were found with enzyme BstYⅠ;only one kind of genotype were found with enzyme TaqⅠ.Polymorphic sites were detected at base position 61,70,94,104,124,150,154,163,173,182,202 and 217 of the exon 2 of BoLA-DRB3 gene.The results of x2 test showed that the polymorphic sites of BstYⅠ and TaqⅠ were in Hardy-Weinberg equilibrium,but the loct of HaeⅢ and RsaⅠ were not in Hardy-weinberg equilibrium.
