γ-亚麻酸治疗兔蒸发过强型干眼的实验研究
Effect of γ-linolenic acid on experimental evaporative dry eye in rabbit
【摘要】 目的观察γ-亚麻酸治疗实验性蒸发过强型干眼的疗效。方法烧灼兔睑板腺开口制成蒸发过强型干眼模型,将30只兔随机分为对照组和治疗组2组。对照组不给予任何药物;治疗组给予γ-亚麻酸灌胃治疗,用药前后定期进行SchirmerⅠ试验、角膜虎红染色,并观察泪膜破裂时间、泪液中白细胞介素-6(IL-6)质量浓度的变化。用药5周后,免疫组织化学方法检测细胞间黏附分子-1(ICAM-1)在角膜、结膜中的表达。结果治疗组兔SchirmerⅠ试验滤纸湿长、泪膜破裂时间较对照组增加(P<0.01)、角膜虎红染色分值下降(P<0.01)、泪液中IL-6质量浓度下降(P<0.01)。ICAM-1在治疗组角膜、结膜上皮无表达或弱表达,在对照组有表达,两者差异有统计学意义(P<0.01)。结论γ-亚麻酸通过对抗细胞因子的作用减轻蒸发过强型干眼眼表的炎症反应,对干眼有治疗作用。
【Abstract】 Objective γ-linolenic acid (GLA) is an essential fatty acids which has extensive physiological action and conspicuous pharmacological action. This experiment was undertaken to observe the effects of γ-linolenic acid on experimental evaporative dry eye in rabbit. Methods Dry eye model was established on thirty rabbits by cauterizing the meibomian gland orifices. The animal was randomly divided into dry eye group and γ-linolenic acid treatment group. Schirmer Ⅰtest,break up time (BUT),Rose Bengal stain and concentration of IL-6 in tear were recorded before and after treatment. Cornea and conjunctival epithelium was examined by immunohistochemical staining to detect the expression of ICAM-1. Results Schirmer Ⅰtest wetting length and BUT time were longer in γ-linolenic acid treatment group than those of control group significantly(12.65±0.65 mm,13.52±1.29 s versus 7.20± 1.23 mm,7.66±1.36 s)(P<0.01). The score of cornea Rose Bengal stain in γ-linolenic acid treatment group (0.15±0.30) decreased significantly in comparison with control group(6.12±0.82) (P<0.01). The concentration of IL-6 in tear was lower in γ-linolenic acid treatment group(112.25±7.15 ng/ml) than control group(244.60±12.60 ng/ml) (P<0.01). The expression of ICAM-1 in cornea and conjunctival epithelium in γ-linolenic acid treatment group was weaker than control group (60.34±10.21 and 54.32±21.45 versus 272.12±46.76 and 302.67±56.28) (P<0.01). Conclusion γ-linolenic acid can alleviate the inflammation in rabbit ocular surface and decrease the concentration of IL-6 in tear,which suggest that γ-linolenic acid is effective in the treatment of dry eye.
