HPLC测定蒙药草乌中药效组分含量
Simultaneous Content Determination of Aconitine, Hypaconitine and Mesaconitine in Aconitum kusnezoffii Reichb. by HPLC
【摘要】 目的:建立以高效液相色谱法同时测定蒙药草乌中乌头碱、新乌头碱、次乌头碱含量的方法。方法:Ex-tend-C18(250mm×4.6mm,5μm),流动相为甲醇-0.3mol·L-1三乙胺(65∶35),流速为0.8mL·min-1,检测波长为235nm,柱温为35℃。结果:乌头碱、次乌头碱、新乌头碱分别在0.35~1.75μg,0.5~2.5μg,1~5μg呈良好的线性关系。结论:本方法简便、准确、分离效果好、线性范围宽、灵敏度高,可用于本品的质量控制。
【Abstract】 Objective: To establish a method for simultaneous content determination of aconitine, hypaconitine and mesaconitine in Aconitum kusnezoffii Reichb.by HPLC.Methods: Extend-C18(250mm×4.6mm,5μm) was used as chromatographic column,methanol-0.3mol · L-1 triethylamine (65∶ 35)as mobile phase. The detection wavelength was set at 235nm,the flow rate at 0.8mL · min-1 and the column temperature at 35℃. Results: The sample sizes of aconitine,hypaconitine and mesaconitine had good linear relationship with their peak areas in the range of 0.35~1.75μg,0.5~2.5μg and 1~5μg,respectively. Conclusion: The established method is convenient and accurate with satisfactory separation effect,wide linear range,and high sensitivity,and can be available for the quality control of Aconitum kusnezoffii Reichb.
