三苯氧胺对人胶质瘤细胞的生长抑制作用
Inhibitory effects of tamoxifen on human glioma cells im vitro
【摘要】 目的探讨蛋白激酶C(PKC)抑制剂三苯氧胺对人胶质瘤细胞SHG-44生长抑制作用。方法SHG-44细胞中PKCα和雌激素受体(ER)的表达用免疫组化,细胞活性分析用台盼兰实验和四唑盐(MTT)比色实验,细胞增殖和凋亡,通过流式细胞术(FCM)进行检测,三苯氧胺作用终浓度为5、2.5、1.25μg/ml;治疗对照为卡氮芥(BCNU),终浓度与三苯氧胺相同。结果SHG-44细胞中PKCα表达阳性,ER表达阴性。三苯氧胺组与空白对照组相比,细胞衰老、脱落、总数减少;细胞活性下降;G0~G1期细胞比例增加,提示细胞增殖受到抑制;凋亡细胞比例增高。与BCNU组相比,在2.5和1.25μg/ml剂量组中出现G0~G1期细胞比例增加;凋亡细胞比例无统计学差异。结论三苯氧胺在体外对SHG-44胶质瘤细胞生长具有明显的抑制作用。
【Abstract】 Objective To study the inhibitory effects of tamoxifen on human SHG-44 glioma cells in vitro.Methods The expression of PKCα and estrogen receptor (ER) was screened by means of immunohistochemistry. The cellular activity was detected by trypan blue test and MTT test. The proliferation and apoptosis were detected by flow cytometer. The concentrations of tamoxifen were 5, 2.5, and 1.25 μg/ml, respectively. Carmustine (BCNU) was used as therapeutic control at the same concentration as tamoxifen.Results PKCα was positive, but ER was negative in SHG-44 cells. Treated with tamoxifen, the cells became aging and shedding. Their total sum was decreased. The cell activity of tamoxifen trealed cells became weaker than that of blank control. The number of cells, prevented in G0-G1 cell cycle, was larger than that in blank control, 2.5 and 1.25 μg/ml BCNU groups. The number of apoptosis cells was larger in tamorifen treated group than that in blank control.Cellular proliferation was inhibited while apoptosis increased. The efficiency of tamoxifen was comparable with BCNU.Conclusion Tamoxifen can inhibit SHG-44 glioma cells in vitro.
