运动训练预防大鼠非酒精性脂肪性肝炎形成实验研究
The Preventive Effect of Exercise on the Formation of Nonalcoholic Steatohepatitis in Rats
【摘要】 目的:探讨游泳训练对大鼠非酒精性脂肪性肝炎形成的影响。方法:30只雄性SD大鼠随机分为4组:对照组(正常饮食12周)、高脂饮食组(高脂饮食12周)、高脂饮食运动组(高脂饮食同时游泳训练12周)和运动组(正常饮食同时游泳训练12周)。12周后处死大鼠,观察各组肝脏组织切片并测试血清以及肝脏各项指标。结果:高脂饮食组大鼠肝脏出现严重的大泡性脂肪变性并有不同程度的炎性细胞浸润,高脂饮食运动组肝脏偶有脂肪变性和炎性细胞,对照组和运动组为正常肝脏组织形态。高脂饮食组的肝脏脂肪变性程度和炎症活动度评分均显著高于对照组,而高脂饮食运动组显著低于高脂组、显著高于对照组。高脂饮食组血清和肝脏游离脂肪酸、血清甘油三酯、肿瘤坏死因子α、肝脏丙二醛含量均显著高于对照组,而高脂饮食运动组显著低于高脂组;高脂饮食运动组血清和肝脏游离脂肪酸、肿瘤坏死因子α、肝脏丙二醛含量与对照组相比无显著性差异;高脂饮食组血清胆固醇显著高于对照组,而肝脏超氧化物歧化酶显著低于对照组,两者与高脂饮食运动组无显著性差异;高脂饮食运动组血清甘油三酯和胆固醇显著高于对照组,而超氧化物歧化酶显著低于对照组。结论:运动训练可以在一定程度上预防非酒精性脂肪性肝炎的形成。
【Abstract】 Objective To study the effect of swimming on the formation of nonalcoholic steatohepatitis(NASH) in rats.Methods Thirty male SD rats were randomly assigned into four groups: control(C,12-week normal diet),high-fat diet(H,12-week high-fat diet),high-fat diet and swimming(HE,high-fat diet plus swimming for 12 weeks),and swimming(E,normal diet plus swimming for 12 weeks) groups.After 12 weeks of swimming,the livers of all rats were observed histologically,and the serum TG,TC,HDL,LDL,FFA,TNFα,liver FFA,MDA,SOD,and content of liver protein were measured.Results Severe macrovesicular steatosis and inflammation were observed in the group H and less steatosis and inflammation appeared in group HE.Normal liver histology was found in group C and E.The score of severity of hepatic steatosis and histological activity index(HAI)in group H was significantly greater than that in group C,and the score of severity of hepatic steatosis and HAI in group HE was lower than that in group H.The serum FFA,TG,TNF-α and liver MDA in group H were significantly higher than that in group C,and serum FFA,TG,TNF-α and liver MDA in group HE were significantly lower than that in group H.There was no significant difference in serum FFA,TNF-α,liver FFA and MDA in group HE and in group C.The serum TC in group H was higher and liver SOD was lower than that in group C,while no significant differences were found between group H and HE.The serum TG and TC in group HE were higher and liver SOD was lower than that in group C. Conclusion Concurrent 12-week swimming could prevent the formation of NASH in rats to some extent.
