CCR3在哮喘豚鼠模型肺和骨髓中的表达及意义

【摘要】 目的观察哮喘豚鼠肺和骨髓组织嗜酸粒细胞趋化因子受体3(CCR3)及嗜酸粒细胞(EOS)不同时相表达的变化,探讨哮喘发病的可能机制。方法用卵白蛋白(OVA)和生理盐水致敏法制备豚鼠哮喘和对照模型,分为正常对照组(N)及哮喘组(A、B、C、D、E),每组8只,于激发后30 min,6 h,12 h,24 h,48 h处死,瑞氏染色计数骨髓涂片和外周血涂片中EOS比例,免疫组织化学技术检测骨髓中CCR3的表达;制备豚鼠肺组织病理标本,苏木精-伊红染色,免疫组化检测肺组织中CCR3的表达。结果①哮喘组骨髓和外周CCR3及EOS表达明显增加;②OVA激发后,骨髓和外周CCR3及EOS表达:30 min变化不明显,6 h,12 h达到峰值,24 h开始下降,48h恢复正常水平;③骨髓的变化早于外周,CCR3的表达早于EOS的增多,且CCR3和EOS呈线性相关。结论哮喘豚鼠肺组织和骨髓之间存在骨髓EOS细胞到循环再到肺组织的通路,CCR3表达增强,为EOS从骨髓快速募集到肺组织提供了可能。

【Abstract】 Objective To explore the pathogenetic mechanism of asthma by tracking the expressions of CCR3 and EOS in lung tissue and bone marrow in guinea-pig asthmatic models.Methods Guinea pigs(GP) were sensitized and challenged with OVA to establish the asthmatic model(groups A,B,C,D and E).The control GP(group N) were instead given injections of sterile saline.The sensitized GP were killed 30 min,6 h,12 h,24 h and 48 h after the challenge,with the control GP killed 12 h after the second injection.Slides prepared from peripheral blood and bone marrow,were stained with Wright′s stain to count the total number and percentage of EOS.The expression of CCR3!in bone marrow and lung tissue was detected by immunohistochemistry.Results ① The total counts of EOS and the expression of CCR3 in bone marrow and lung tissue were significantly higher in allergic-asthma models than in controls.②After the challenge with OVA,the levels of CCR3 and EOS remained in the normal range at 30 min,began to rise and reached the peak at 6 h and 12 h,found to be decreased at 24 h and returned to normal by 48 h.③The bone marrow changes appeared earlier than that in the periphery.The expression of CCR3 was earlier than the recruitment of EOS and they two were of linear correlation.Conclusion There is a pathway for the bone marrow EOS stem cells to move to the circulating blood to the lung tissue.The increase of CCR3 expression would enable the recruitment of EOS from bone marrow to the lung.
