载脂蛋白CⅡ、载脂蛋白CⅢ含量和脂蛋白脂肪酶、肝脂肪酶活性在大鼠脂肪肝模型中的变化

【摘要】 目的观察载脂蛋白CⅡ、载脂蛋白CⅢ和脂蛋白脂肪酶、肝脂肪酶在大鼠脂肪肝模型中的变化,探讨大鼠脂肪肝发病机理。方法高脂肪、高胆固醇饮食饲养法造脂肪肝胰岛素抵抗大鼠模型;检测马来酸罗格列酮干预前后载脂蛋白CⅡ、载脂蛋白CⅢ含量和脂蛋白脂肪酶、肝脂肪酶活性的变化;用双抗体夹心ELISA法检测脂肪肝大鼠血浆载脂蛋白CⅡ、载脂蛋白CⅢ含量,用酶法检测脂肪肝大鼠脂蛋白脂肪酶、肝脂肪酶活性。结果脂肪肝大鼠的血浆载脂蛋白CⅡ含量较正常大鼠降低,载脂蛋白CⅢ含量较正常大鼠升高,脂蛋白脂肪酶、肝脂肪酶活性较正常大鼠降低;经马来酸罗格列酮干预治疗后,脂肪肝大鼠的血浆载脂蛋白CⅡ含量升高,载脂蛋白CⅢ含量降低,脂蛋白脂肪酶、肝脂肪酶活性均升高。结论胰岛素抵抗使载脂蛋白CⅡ含量减少、载脂蛋白CⅢ增多,致脂蛋白脂肪酶、肝脂肪酶活性降低,而胰岛素增敏剂可以通过改变载脂蛋白CⅡ、载脂蛋白CⅢ含量来提高脂蛋白脂肪酶,肝脂肪酶活性。

【Abstract】 Objectives To observe the variations of plasmic apolipoprotein-CⅡ、apolipoprotein-CⅢ content and the plasmic activity of lipoprotein lipase and hepatic lipase and to investigate the the pathogenesis of nonalcoholic fatty liver disease of rats.Methods The rats fed with hypsi-fat and hypsi-cholesterol forage to establish nonalcoholic fatty liver model resistant to insulin.The variations of plasmic apolipoprotein-Ⅱ and apolipoprotein-Ⅲ content and the activity of plasmic lipoprotein lipase and hepatic lipase before and after the therapy of Rosiglitazone maleate were detected using enzyme-linked.immunosorbent-assay and enzyme method respectively.Results Compared with the normal rats,the plasmic apolipoprotein-CⅡ content and activity of lipoprotein lipase and hepatic lipase of nonalcoholic fatty liver disease rats were lower,while the plasmic apolipoprotein-CⅢ content were higher.Conclusions Insulin resistance can increase the plasmic apolipoprotein-CⅢ content in nonalcoholic fatty liver disease rats and decrease the plasmic apolipoprotein-CⅡ content as well as the activity of plasmic lipoprotein lipase and hepatic lipase;euglycemic agent can raise the plasmic activity of lipoprotein lipase and hepatic lipase by changing the content of pasmic apolipoprotein-CⅡ and apolipoprotein-CⅢ in nonalcoholic fatty liver disease rats.
