花萼海绵体诱癌素A诱导早熟染色体凝集的方法学探讨

【摘要】 目的探讨用花萼海绵体诱癌素A(Calyculin A)诱导的外周血淋巴细胞早熟染色体凝集(PCC)的最优化方法。方法采用х2检验,对影响Calyculin A诱导的外周血淋巴细胞PCC的3个主要影响因素(Calyculin A的终浓度、处理时间、淋巴细胞培养时间)进行两两比较分析。结果随着Calyculin A终浓度的升高,分裂指数亦升高,10、50nmol/L两组间差异有显著性(P<0.05),50、100nmol/L两组间差异无显著性(P>0.05);随着CalyculinA处理时间的延长,分裂指数亦逐渐升高,各组间差异均有显著性(P<0.05)。结论Calyculin A终浓度为50nmol/L,处理时间为2h,淋巴细胞培养时间为48h的实验组合可以获得较高的分裂指数,远远高于秋水仙素常规组的分裂指数。

【Abstract】 Objective To explore a optimal method for prematurely condensed chromosome(PCC)induced by calyculin A peripheral blood lymphocyles.Methods х2-test was used to compare and analyze three mainly influencing factors of PCC induced by calyculin A.Results With the rise of final concentration of calyculin A,divided index increased.Significant difference was shown between the groups of 10 nmol/L and 50 nmol/L groups(P<0.05),but not between 50 nmol/L and 100 nmol/L groups(P>0.05).With the delay of disposed time,divided index also increased,showing significant differences(P<0.05).Conclusion When the final concentration reached to 50 nmol/L,disposed time was set to 2 hours and lymphocytes cell were cultivated for 48 hours,high divided index can be attained,which is much higher than that acquired by routine method with colchicine.
