HPLC法测定红穿破石中大黄素的含量

【摘要】 目的建立红穿破石中大黄素含量的高效液相色谱测定方法。方法选用EclipseXDB-C18色谱柱(4.6mm×150mm,5μm),以甲醇-0.1%磷酸溶液(75∶25)为流动相,检测波长为285nm。结果大黄素在0.0063~0.063μg范围内呈良好的线性关系(r=0.9999),平均回收率为102.0%,RSD=1.58%(n=6)。结论本法简便,准确,可用于红穿破石中大黄素的含量测定。

【Abstract】 Objective To establish a HPLC method for determining emodin in Radix et Caulis Ventilaginis leiocarpae. Methods Stationary phase was Agilent Eclipse XDB-C18 (4.6 mm×150 mm, 5 μm), mobile phase was methanol-0.1%phosphoric (75∶25), detection wavelength was 285 nm, flow rate was 1.0 mL/min and column temperature was 30 ℃. Results There was good linearity in the range of 0.006 3~0.063 μg for emodin (r=0.999 9). The average recovery of emodin was 102.0% with RSD=1.58%. Conclusion This method is rapid and accurate. It can be applied to control the quality of Radix et Caulis Ventilaginis leiocarpae.
