青刺果多糖对糖尿病小鼠肺组织病理变化的影响

【摘要】 为了观察青刺果多糖对四氧嘧啶诱导的糖尿病小鼠肺组织病理变化的影响,探讨其对糖尿病小鼠肺脏损伤的保护作用,对腹腔一次性注射四氧嘧啶复制糖尿病模型.造模成功后,将实验动物分为模型组和青刺果多糖治疗组(300mg/kg),另设正常对照组(生理盐水).实验小鼠分别于给药2周末和4周末随机抽取处死,取肺脏固定于4%的多聚甲醛中,制作病理切片,进行HE和Gomori镀银染色,光镜下观察.结果表明:光镜下,HE染色可见模型组小鼠肺泡间隔增宽,支气管周围和肺泡间隔大量炎性细胞浸润,毛细血管基膜增厚,含铁血黄素沉积明显,Gomori镀银染色可见支气管、肺泡间隔和小血管周围网状纤维和胶原纤维含量增多.青刺果多糖治疗组,肺泡间隔和肺泡腔有轻微的淤血,支气管周围纤维细胞轻微增生和少量炎性细胞浸润,支气管、肺泡间隔和小血管周围有少量网状纤维和胶原纤维,并得出青刺果多糖对糖尿病小鼠肺脏损伤具有保护作用.

【Abstract】 To study the effects of polysaccharides from Prinsepia utilis Royle on the pathological changes induced by alloxan in the lungs of diabetic mice and to investigate their protective mechanism.Alloxan was administered intraperitoneally to establish diabetic models in mice.After the modeling,the experimental animals were divided into the model group(physiological saline),polysaccharides from Prinsepia utilis Royle treatment group(300mg/kg)and normal control group(physiological saline).Two and four weeks after administration,the mice were randomly sacrificed,and their lings were removed and fixed into the 4% paraformaldehyde.Their pathological sections were silver-stained(hematoxylin and eosin staining and gomori silver staining methods)and observed under a light microscope.In the model group,we observed enlargement of alveolar septum,inflammatory cell infiltrate in bronchus and alveolar septum,thickened pulmonary capillary basal lamina and hemosiderin deposition obviously with hematoxylin and eosin staining,increasing content of reticular fibers and collagenous fibers in bronchus,alveolar septum and small vessels with Gomori silver staining under the light microscope.After treatment with polysaccharides from P.utilis Royle,slight congestion appeared in alveolar septum and alveolar space,little fibrocyte accrementite and small inflammatory cell infiltrate in bronchus surrounding,small amounts of reticular fibers and collagenous fibers in bronchus,alveolar septum and small vessels.We got polysaccharides from P.utilis Royle can protect the lungs of the diabetic mice.
