Ccnd1反义寡核苷酸脂质体对囊外摘出术后大鼠晶状体上皮细胞增殖的抑制作用
Inhibiting effect of Ccnd1 antisense oligonucleotides liposomes on proliferation of lens epithelial cells after extracapsular lens extraction in rats
【摘要】 目的探索Ccnd1反义寡核苷酸(Ccnd1antisense oligonucleotides,Ccnd1-ASON)脂质体对晶状体囊外摘出术(extracapsular extraction of lens,ECLE)后大鼠晶状体上皮细胞(lens epithelial cells,LEC)增生的抑制作用。方法48只SD大鼠随机分为3组,双眼行ECLE,术中前房内分别注入Ccnd1-ASON脂质体溶液(反义组)、Ccnd1基因正义寡核苷酸(Ccnd1sense oligonucleotides,Ccnd1-SON)脂质体溶液(正义组)、无药脂质体溶液(无药组)50μL;每组随机取4只分别于术后即刻、1d、7d、14d处死,实时荧光定量PCR检测LECCcnd1表达变化,组织病理学检测单位面积LEC数量,免疫组织化学检测LEC周期蛋白(cyclin)D1表达,应用方差分析进行统计学处理。结果术后1d、7d、14d,Ccnd1表达量反义组分别为0.1415、0.2743、0.3406,低于正义组(0.7861、1.1623、0.7537)和无药组;晶状体组织单位面积(mm2)LEC数量反义组分别为633±65、1636±212、2440±359,低于正义组(808±73、2476±349、3115±316)和无药组(845±78、2242±303、3133±331)(P<0.05),正义组和无药组之间无明显差异(P>0.05);晶状体组织单位面积(mm2)Cyclin D1阳性细胞积分光密度值反义组分别为6.18±0.67、11.20±1.66、17.38±3.00,低于正义组(7.74±0.88、16.03±2.40、24.56±2.49)和无药组(7.60±0.66、16.17±2.03、25.93±2.21)(P<0.05),正义组和无药组之间无明显差异(P>0.05)。结论Ccnd1-ASON前房注射能有效降低大鼠ECLE后LEC Ccnd1和Cyclin D1表达,抑制LEC增殖,减轻后囊膜混浊。
【Abstract】 Objective To evaluate the inhibiting effect of Ccnd1 antisense oligonucleotides(Ccnd1-ASON)liposomes on the proliferation of lens epithelial cells(LEC)after extracapsular lens extraction(ECLE)in rats.Methods Fourty-eight SD rats were divided into 3 groups at random and all received ECLE at both eyes.Ccnd1-ASON liposomes,Ccnd1 sense oligonucleotides(Ccnd1-SON)liposomes and liposome without any drugs were respectively injected into anterior chamber of different groups.Four rats of each group were sacrefied at 0 hour,1st,7th,and 14th day after surgery and the eyes were enucleated.The expression of Ccnd1 in LEC was detected using quantitative real-time polymerase chitin reaction.The numbers of LEC in every square were measured with light microscopy and the expression of cyclin D1 in LEC examined by immunohistochemical technique.Variance analysis was used to compare the number of proliferative LEC and the integrated optical density of cyclin D1 positive LEC.Results At the time after 1st,7th,and 14th day of operation,the expression quantity of Ccnd1 in group of Ccnd1-ASON(0.141 5,0.274 3,0.340 6)was less than the group of Ccnd1-SON(0.786 1,1.162 3,0.753 7)and the control;The numbers of LEC per mm2 in group of Ccnd1-ASON(633±65,1 636±212,2 440±359)was less than the group of Ccnd1-SON(808±73,2 476±349,3 115±316)and the control(845±78,2 242±303,3 133±331)(P<0.05)while the later two groups got no difference;the integrated optical density of cyclin D1 positive LEC per mm2 in group of Ccnd1-ASON(6.18±0.67,11.20±1.66,17.38±3.00)was less than the group of Ccnd1-SON(7.74±0.88,16.03±2.40,24.56±2.49)and the control(7.60±0.66,16.17±2.03,25.93±2.21)(P<0.05)while the later two groups got no difference.Conclusion Anterior chamber injection of Ccnd1-ASON liposomes can inhibit the proliferation of LEC and reduced posterior capsule opacification in some degree.
