原发性开角型青光眼家系永生细胞株的建立与保存
Establishment and preservation of immortal lymphoblastoid cell lines of the primary open angle glaucoma pedigree in Chongqing
【摘要】 目的建立并保存我国重庆地区原发性开角型青光眼(primary open angle glaucoma,POAG)大家系患者及其亲属永生淋巴细胞株,为进一步深入研究该家系奠定良好基础。方法采用EB病毒(Epstein-barr virus,EBV)转化外周血淋巴细胞同时加入环孢霉素A(cyclosporin A,CsA)法,建立该家系永生细胞株。结果建株成功率达到92.3%,对已建立的永生细胞株经复苏和冻存的成功率为100.0%。结论成功建立并保存了这一具有重要研究价值的家系遗传资源,为在细胞和分子水平上进一步开展POAG的基础研究提供了宝贵资源。
【Abstract】 Objective To further explore the pathologic mechanisms of primary open angle glaucoma(POAG)and establish and preserve the immortal lymphoblastoid cell lines in a large POAG pedigree of Chongqing.Methods The immortal lymphoblastoid cell lines were established by Epstein-barr virus(EBV) transformation of peripheral blood lymphocytes and addition of cyclosporine A.In this present study,39 immortal cell lines of the large POAG pedigree were established,including 11 POAG patients and 28 unaffected relatives (21 males and 18 females).Results The immortal lymphoblastoid cell lines has been proved to be successful in above 92.3% of blood samples and in almost 100% for rejuvenate cells.Conclusion The successfully established immortal lymphoblastoid cell lines are important for cytogenetics and molecular biology study of POAG.
