超临界流体膨胀减压过程制备水溶性药物超细微粒
Preparation of water-soluble drug microparticles by supercritical fluid expansion depressurization process
【摘要】 利用自建的超临界流体膨胀减压过程实验装置,以不同水和乙醇配比的混和溶剂制备了扑热息痛,四环素和头孢羟氨苄超细微粒,并研究了不同溶剂配比对过程实现的影响。结果表明,用超临界膨胀减压过程可以成功制备水溶性药物超细微粒,制得的大部分微粒为球形或近球形,粒径处于1~3μm之间,粒径分布均匀;以纯水为溶剂时,过程的可行性较差,喷嘴堵塞现象严重;在初始溶液中加入一定配比的乙醇,会使过程进行顺利,制备的微粒的球形度更好,粒径更小,分布更均匀。
【Abstract】 The microparticles of paracetamol,tetracycline and cefadroxil are prepared successfully from water-ethanol mix-solvent with different volume ratios by supercritical fluid expansion depressurization(SFED) process.The feasibility of the process with different solvent volume ratios is investigated.The results show that the water-soluble drug microparticles can be prepared successfully by SFED process.With pure water as the solvent,the process performs badly and the blockage of nozzle occurrs frequently.With the mixture of water and ethanol at a certain volume ratio as the solvent,the process can be conducted favorably.The blockage of nozzle can be avoided and the particles prepared have better spherical shape,smaller size and narrower size distribution.
