大黄素、黄芪多糖在体外对乙型肝炎病毒的抑制作用

【摘要】 目的体外观察大黄素、黄芪多糖单用与联合应用抗HBV的作用。方法以HepG2.2.15细胞为研究模型,选用干扰素α、拉米夫定为阳性对照药物,观察培养液中加入不同浓度大黄素、黄芪多糖及两药联合应用抗HBV作用的效果。采用Taqman荧光定量PCR检测细胞上清HBV DNA,ELISA检测培养上清液HBsAg、HBeAg水平的变化。采用MTT法观察它们对细胞生长的影响。结果大黄素的3个剂量组(12.5、25.05、0.0 mg/L)和所观察的时间点(3、69、d)对HBV DNA、HBsAg、HBeAg表现出剂量与时间依赖的抑制作用(P<0.05)。大黄素对HBV DNA抑制的半数抑制浓度为21 mg/L,对HepG2.2.15细胞半数细胞毒性浓度为40.66 mg/L,治疗指数为1.94。同时发现黄芪多糖各剂量组对HBV无明显的抑制作用(P>0.05)。大黄素、黄芪多糖联合应用在疗效方面无明显的交互作用(P>0.05)。结论大黄素在体外对HBV具有一定的抑制作用,黄芪多糖无明显抑制HBV的作用,两药联用无明显协同抗HBV的作用。

【Abstract】 Objective To investigate the effects of emodin,astragalus polysaccharides(APS) on hepatitis B virus(HBV) in vitro.Methods Human hepatoma G2.2.15 cell line stably expressed hepatitis B virus particles in culture.The cells were exposed to different concentrations of emodin,APS,interferon α,and lamivudin,respectively.Real-time PCR was applied to quantify extracellular HBV DNA and ELISA was applied to assess HBsAg and HBeAg.MTT assay was used to evaluate the cytotoxicity of drugs.Results The results showed that exposure of HepG2.2.15 cells to emodin resulted in a time-and concentration-dependent inhibition of HBV DNA replication and HBsAg,HBeAg secretion(P<0.05).IC50 to HBV DNA was 21mg/L,CC50 to HepG2.2.15 cells was 40.66mg/L,and the treatment index(TI) was 1.94.APS could not restrain HBV DNA,HBsAg and HBeAg obviously in vitro(P>0.05).The interaction was not significant between emodin and APS(P>0.05).Conclusion Emodin had time-and concentration-dependent inhibitory effects to HBV in vitro in some extent;APS could not inhibit HBV obviously in vitro;Co-administration of emodin and APS had not significant interaction in restraining HBV in vitro.
