红景天苷对全脑缺血再灌注大鼠皮层nNOS及核酸的影响

【摘要】 【目的】探讨红景天苷(salidroside)对全脑缺血再灌注(cerebral ischemic reperfusion,CIR)大鼠大脑皮层一氧化氮合酶(nitric oxide synthase,NOS)及核酸表达的影响。【方法】用改良的Pulsinelli 4-血管阻断(4-VO)法,复制大鼠急性全脑缺血(30 min)再灌注(60 min)损伤模型。尼克酰胺腺嘌呤二核苷酸磷酸黄递酶(NADPHd)组化染色法检测神经元型一氧化氮合酶(nNOS),吖啶橙(AO)染色观察DNA、RNA的变化。【结果】CIR模型组大鼠脑内nNOS表达增加,DNA和RNA荧光强度(反映DNA和RNA含量)明显减弱。灌胃给予红景天苷后可抑制CIR大鼠大脑皮层nNOS的表达,DNA和RNA荧光强度增强。【结论】红景天苷可抑制CIR导致的大鼠大脑皮层nNOS的表达,对DNA和RNA有保护作用。

【Abstract】 【Objective】To explore the effect of salidroside on the nitric oxide synthase and nucleic acid in the cerebral cortex of acute global cerebral ischemia-reperfusion(CIR) injury in rats.【Methods】Improved Pulsinelli’s 4-vessel occlusion(4-VO) method was used to set up the rat models of acute global cerebral ischemia-(30 min) reperfusion(60 min).nNOS expression neurons in cerebral cortex was determined by NADPHd histochemical method.The acridine orange(AO)method was carried out to observe the changes of DNA and RNA in cerebral cortex.【Results】nNOS expression neurons in the cerebral cortex of CIR rats showed obvious increase after 30 min of ischemia and 60 min of repufusion.Intragastric administration of salidroside inhibited the increase of nNOS expression neurons significantly.Fluorescent intensity(reflexing content of DNA and RNA) was unclear and weaker obviously.In cerebral cortex of CIR rats,intragastric administration of salidroside could restrain the cut-off of DNA and RNA chain and the fluorescent intensity was clear was enhanced.【Conclusions】Salidroside represses the expression of nitric oxide synthase in CIR injured rats.and has the protective effect on DNA and RNA and relieves the damage of global cerebral ischemia-reperfusion.
