内置鞘管法建立小鼠颈动脉粥样硬化模型
Establishment of mice carotid artery atherosclerosis model with inlay sheathing canal way
【摘要】 目的:探求一种新的动脉粥样硬化模型建立方法。方法:40只雌性C57BL/6小鼠随机分为造模组和对照组,造模组通过左侧颈动脉外放置鞘管方法建立颈动脉狭窄模型,两组小鼠均饲以高脂饮食12周,分别留取造模前后血清进行生化检测,颈动脉标本进行苏木精-伊红(HE)和细胞间粘附分子-1(ICAM-1)染色。结果:12周后两组小鼠总胆固醇(TC)和低密度脂蛋白胆固醇(LDL-C)均较造模前有明显增加,巨噬细胞集落刺激因子(M-CSF)造模组明显高于对照组,造模组ICAM-1染色呈阳性,HE染色可见造模组明显动脉粥样硬化形成。结论:通过内置鞘管法可成功建立小鼠颈动脉粥样硬化模型。
【Abstract】 Objective: To find a new way to establish animal model of atherosclerosis.Methods: 40 female C57BL/6 mice were randomly divided into model group and control group.Sheathing canal was placed on left carotid artery in model group.Each group was given high-cholesterol diet for 12 weeks.At the end both sides of carotid arteries were taken and hematoxylin and eosin-stained sections,ICAM-1 stained were used for morphometric analysis.The serum level of cholesterol(TC),low-density lipoprotein cholesterol(LDL-C),macrophage colony stimulating factor(M-CSF) were measured.Results: In two serum groups level of TC and LDL-C increased significantly after 12 weeks than before,M-CSF serum level was much higher in model group than that in control group.the early atherosclerosis formation on the left side was observed and ICAM-1 stained was positive in the model group.Conclusion: Inlay sheating canal way can be successful used to establish mice carotid artery atherosclerosis model.
