【摘要】 目的观察类风湿性关节炎(rheumatoid arthritis,RA)患者外周血和滑膜液中树突状细胞(dendritic cell,DC)亚群(DC1、DC2)变化的临床意义及与T辅助细胞亚群(Th1、Th2)间的关联。方法采用免疫荧光染色技术标记RA患者外周血单个核细胞和关节滑液中单个核细胞表面DC1和DC2组合标志、CD4分子和细胞内IFN-γI、L-4分子,用流式细胞术分析标记结果。结果实验发现虽然RA患者外周血DC亚群含量与对照组无明显差异,但关节液中DC1数量较RA和对照组外周血组明显增高,而DC2含量则明显低于RA和对照组外周血组(P<0.05~0.01)。RA外周血和关节液中Th1细胞数量和Th1/Th2比值较对照组外周血升高(P<0.01)。DC1数量和DC1/DC2比值与Th1含量有正相关关系(P<0.01~0.05)。结论RA患者的病程中存在DC亚群的异常,这种异常改变可能导致Th亚群比例的失衡及疾病的活动。更多还原

【Abstract】 Objective To investigate the alteration of the subsets of dendritic cells(DC) from peripheral blood(PB) and synovial fluid(SF) in patients with rheumatoid arthritis(RA) and its correlation with the subsets of T helper cells.Methods Immunofluorescence staining technique were used to label the combined marker for DC1,DC2 on the cell surface and also to label CD4 and intracellular marker for IFN-γ、IL-4 of PBMC and SFMC from 37 RA patients.The labeled cells were then analyzed by flow cytometry.Results Although DC subset showed no obvious difference between RAs’ and controls’ PB,DC1 level increased significantly in RA SF compared with that in RA and controls PB(P<0.05～0.01).The quantity of DC2 subset in RA SF was however proved to be lower than those in RA and controls’ PB(P<0.05～0.01).The high frequency of Th1 and ratio of Th1/Th2 in both RA SF and PB were observed as compared with controls’ PB(P<0.01).DC1 level and DC1/DC2 ratio were correlated with the number of Th1 subset(P<0.01,0.05).Conclusion The level of DC subset is abnormal during the course of rheumatoid arthritis that probably result in the unbalance of T helper subset and disease activity.更多还原
