大黄素对泼尼松致大鼠脂代谢异常的预防作用研究

【摘要】 目的观察不同浓度大黄素对泼尼松致大鼠脂代谢异常的预防作用。方法3月龄大鼠给予泼尼松和药物90d。观察血清生化指标、器官指数和骨髓基质细胞体外的成脂能力。结果给予泼尼松和药物90d后大鼠高密度脂蛋白胆固醇降低,骨髓基质细胞体外成脂能力增强。大黄素90mg·kg-1·d-1和大黄素270mg·kg-1·d-1可升高高密度脂蛋白胆固醇和骨髓基质细胞体外成脂能力。结论大黄素对泼尼松引起的脂代谢异常有预防作用。

【Abstract】 Objective To determine whether emodin can completely prevent dyslipidemia in rats treated with prednison. Methods Thirty-five Sprague-Dawley male rats at age of 3 months were divided into five groups and treated for 90 days as follows: intact rats were given with vehicle as control; Prednisone was given orally to the rats at dose of 3.0 mg/kg/d as the model group; Emodin at dose of 90mg/kg/d or 270 mg/kg/d were plus to the prednison treated rats and Integrated medicine (Calcium 375 mg/kg/d+VitD3 250 U/kg/d ) were plus to the prednison treated rats. The blood serum was separated for testing biochemistry indices. the MSCs were isolated by density-gradient centrifugation and cultured.Results Rats treated with GC decreased serum HDL-Cholesterol and significantly increased serum sulfur compared with vehicle controls. Treatment with emodin markedly increased in the serum HDL-Cholesterol level and decreased serum sulfur. Emodin prevented adipogenic differentiation of MSCs from prednisone-treated rats.Conclusion Emodin shows effect of prevention against Glucocorticoid -induced dyslipidemia in rats.
