肺癌患者射线诱发的淋巴细胞遗传损伤及ATM蛋白表达的研究

【摘要】 目的观察肺癌患者射线(IR)诱发的淋巴细胞遗传损伤及与ATM(ataxia telangiectasia mutated)蛋白表达的关系。方法淋巴细胞采自22例肺癌患者和22例对照。每个样本分为两部分:非辐射样本和辐射样本(3Gy X-ray)。用彗星试验和微核(MN)试验检测肺癌患者和对照组淋巴细胞自发的和IR诱发的遗传损伤。用Western blotting试验测定肺癌患者和对照组淋巴细胞的ATM蛋白表达水平。结果肺癌患者彗星试验的平均尾相(MTM)和微核试验的微核率(MNR)基础值分别为0.86‰和11.41‰,明显高于对照组的0.64‰和6.77‰(P值分别<0.05和<0.01)。IR诱发的平均MTM和MNR肺癌患者分别为1.23‰和77.64‰,明显高于对照组的0.71‰和66.05‰(P值分别<0.05和<0.01)。肺癌患者的ATM蛋白表达为0.64,明显低于对照组的1.71(P<0.01)。结论肺癌患者淋巴细胞对射线的敏感性明显高于对照组,但ATM蛋白的表达明显低于对照组。

【Abstract】 Objective To observe the genetic damage induced by ionizing radiation(IR) and its relationship with the ataxia telangiectasia mutated(ATM) protein expression levels in peripheral lymphocytes of lung cancer patients.Methods The peripheral lymphocytes were from 22 lung cancer patients and 22 controls.Each sample was divided into two parts: non-irradiated sample and irradiated sample which was exposed to 3 Gy X-ray.The spontaneous and IR-induced genetic damage in both lung cancer patients and controls was detected with comet assay and micronucleus(MN) assay,and the ATM protein expression levels of non-irradiated samples in lung cancer patients and controls were measured by Western blotting.Results The baseline values of average mean tail moment(MTM) and micronucleus rate(MNR) in lung cancer patients were 0.86‰ and 11.41‰,respectively,which was significantly higher than those(0.64‰ and 6.77‰) of controls(P<0.05 for MTM,P<0.01 for MNR).The IR-induced average MTM and MNR in lung cancer patients were 1.23‰ and 77.64‰,respectively,which was also significantly higher than those(0.71‰ and 66.05‰) of controls(P<0.05 for MTM,P<0.01 for MNR).ATM protein expression level in lung cancer patients was 0.64,which was significantly lower than that(1.71) in controls(P<0.01).Conclusion The radiosensitivity of lymphocytes measured by comet assay and MN assay in lung cancer patients is significantly higher than that in controls,but ATM protein expression level in lung cancer patients significantly lower than that in controls.
