【摘要】 目的:建立一种痢疾志贺菌、沙门菌和单增李斯特菌的多重PCR诊断方法。方法:根据伤寒沙门菌VipR基因、痢疾杆菌侵袭性质粒抗原H(ipaH)基因和单增李斯特菌溶血素hlyA基因序列作为目的基因片段痢疾志贺菌、沙门菌和单增李斯特菌的特异性抗原基因序列,分别设计一对引物,优化各种工作条件,建立一种多重PCR诊断方法。结果:本法特异性好,敏感性可达104拷贝,整个实验过程在3 h～5 h内完成。结论:建立的方法在理论和实际应用上都有优越性,且一次可检测出三种病原菌,可用于临床诊断。更多还原

【Abstract】 Objective To develope a method for detection of Salmonella typhimurium,Bacillus dysenteriae and Listeria monocytogenes by multiplex PCR.Methods Characteristic genes such as VipR,ipaH and hlyA were chosen as the target genes for duplex PCR system.Many factors of duplex PCR were optimized.Results The specificity was satisfactory and the sensitivity was up to 104 copies of plasmids in the system.The whole test can be completed in 3h～5h.Conclusion The method has the superiority in theory and practical application,and also examine three three pathogens once.It can be used in clinical diagnosis.更多还原
