【摘要】 目的探讨端粒酶逆转录酶(hTERT)、c-myc、cyclin D1基因在胶质瘤中的表达与预后的关系。方法收集56例手术切除的胶质瘤标本,应用原位杂交技术和免疫组化S-P法分别检测胶质瘤组织中hTERT和c-myc、cyclin D1基因的表达;6例正常脑组织作为对照。并对检测结果与随访结果进行综合分析。结果56例胶质瘤中,30例表达hTERT,阳性率为53.6%;33例表达c-myc基因,阳性率为58.9%;32例表达cyclin D1基因,阳性率为57.1%。正常脑组织不表达hTERT、c-myc、cyclin D1。随着胶质瘤分级的升高,hTERT和c-myc、cyclin D1基因表达增强(P<0.05)。hTERT与c-myc、cyclin D1基因的表达两两成正相关关系(P<0.05),三者的表达均与患者生存时间成负相关关系(P<0.01)。结论hTERT、c-myc、cyclin D1基因的过表达与胶质瘤细胞的恶性变和过度增殖有关,三者联合检测可作为临床判断胶质瘤患者预后的重要指标。更多还原

【Abstract】 Objective To investigate the expressions of hTERT,c-myc and cyclin D1 in gliomas and discuss its relation to prognosis.Methods In situ hybridization and S-P immunohistochemistry were used to detect expressions of hTERT,c-myc and cyclin D 1 in 56 specimens of human gliomas respectively.Six specimens of normal brain tissues were used to be the controls.Aggregate analysis was used to the results of expreriment and follow-up.Results The positivity rate of hTERT,c-myc and cyclin D1 in 56 gliomas was 53.6%(30/56),58.9%(33/56) and 57.1%(32/56)respectively.There was no expression of hTERT,c-myc and cyclin D1 in the normal brain tissue.The expression of hTERT,c-myc and cyclin D1 strengthened with the rise in the grading of glioma (P<0.05).The ex- pression of hTERT,c-myc and cyclin D1 was positively correlated with each other(P<0.05),but nega- tively correlated with surviving time of the patients(P<0.01).Conclusion The overexpression of hTERT,c-myc and cyclin D1 is related to the malignant change and overproliferation of glioma cells. Their combinied ditecting may be an important index of judging the prognosis of patients with glioma.更多还原
