人脑胶质瘤wnt1、β-catenin、gsk-3β的表达
Expression of wnt1,β-catenin,gsk-3βin the Human Gliomas
【摘要】 目的探讨wnt1信号转导通路主要表达蛋白wnt1、β-catenin、gsk-3β在人脑胶质瘤的表达,并研究其与细胞增殖的相关性。方法利用免疫组化方法检测wnt1、β-catenin、gsk-3β在正常脑组织、低/高级别胶质瘤细胞中的表达。结果正常脑组织中wnt1、β-eatenin、gsk-3β表达均为阴性或弱阳性;wnt1、β-catenin表达均随胶质瘤级别增高而表达增高,且表达均与肿瘤级别成正相关(P<0.01),wnt1、β-catenin的表达水平彼此间均呈正相关(P<0.01);而gsk- 3β随胶质瘤级别增高而表达降低,表达与组织分级成负相关(P<0.01)。结论wnt1和β-catenin表达异常增加及gsk- 3β表达异常减少均是胶质瘤发生、发展的重要因素。
【Abstract】 Objective To detect the expression of wntl,β-catenin,gsk-3βin gliomas and their rela- tionship in gliomas.Methods Four tissue chips comprised of 6 cases of normal brain tissue and 94 cases of glioma tissues.Immunohistochemistry was employed to detect the expression of wntl,β-catenin, gsk-3β.Results The increasing expression of wntl,β-catenin increased was associated with the de- gree of malignancy of tumors,however gsk-3βreduced.The difference of positivity rate and expression level between the low-grade and high-grade tumors were statistically significant.In addition,the expres- sion of wntl andβ-catenin,was correlated well eath other(P<0.01).Conclusion Up-regulation of wnt1 andβ-catenin expression can be used as an important histological sign for the diagnosis and identifica- tion of human tumor.β-catenin overexpression and downward gsk-3βwould be related to gliomas proli- feration.
