谷氨酸对PC12细胞中磷酸化肌细胞增强因子2A蛋白表达的影响
Effects of Glutamate on the Expression of Phosphorylated Myocyte Enhancer Factor 2A Protein in PC12 cells
【摘要】 目的研究兴奋性氨基酸谷氨酸对PC12细胞中磷酸化肌细胞增强因子2A(P-MEF2A)蛋白表达的影响与意义。方法体外培养PC12细胞,设立正常对照组,0.1、1.0、10.0 nmol/L谷氨酸处理组。Western blot测定P-MEF2A与突触素1的蛋白表达量,并进行2种蛋白表达相关性分析。结果谷氨酸诱导PC12细胞中P-MEF2A蛋白表达升高,呈剂量依赖性(F=51.54 P<0.01)。10.0 nmol/L谷氨酸处理24 h后,P-MEF2A蛋白的表达与突触素1的表达量呈显著负相关(r=-0.788 P<0.01)。结论谷氨酸诱导PC12细胞P-MEF2A表达升高,并与学习记忆调控相关基因突触素1的表达下调有关。
【Abstract】 Objective To investigate the effects of glutamate on the expression of phosphorylated myocyte enhancer factor 2A(P-MEF 2A) protein in PC12 cells.Methods PC12 cells were incubated and were divided into normal group,0.1 nmol/L,1.0 nmol/L,10.0 nmol/L glutamate treated group.Western blot was used to detect the protein levels of P-MEF2A and synapsin-1.Results Glutamate induced the increased of P-MEF2A protein levels in a dose-depended way(F=51.54 P<0.01).After PC12 cells were exposed to 10.0 nmol/L glutamate for 24 h,the expression levels of P-MEF2A protein were significantly inverse correlated with synapsin-1 protein levels(r =-0.788 P<0.01).Conclusions Glutamate induced the increase of P-MEF2A in PC12 cells.And it might be correlated with the decrease of synapsin-1 which is related with learning and memory.
