【摘要】 目的采用一种独特的抗体匹配研究建立检测人体外周血妊娠相关血浆蛋白A酶免测定方法。方法以抗妊娠相关血浆蛋白A(PAPP-A)为捕获抗体包被固相载,结合标本中的抗原成分;以抗嗜伊红主要碱性蛋白前体(ProMBP)抗体为检测抗体,标记辣根过氧化物酶,经酶催化底物显色,定量测定人体外周血妊娠相关血浆蛋白A。结果采用过碘酸钠法与离心超滤纯化相结合的方法成功制备抗ProMBP酶标记抗体;所建立的方法方法的灵敏性为0.06mIU/ml,精密度测试平均变异系数为4.59%,特异性实验显示本文所建立方法具有较高的特异性。与国外同类方法相比,差异无显著性。结论采用独特的抗体配对所建立的人体外周血PAPP-A固相酶免疫测定方法灵敏、可靠,与国外同类试剂之间检测结果无显著性差异。更多还原

【Abstract】 Objective To develop a Enzyme immunoassay.assay for testing the concentration of pregnancy-associated plasma protein A,a particular paired antibody is used.Methods A monoclonal antibody specific for PAPP-A was fixed to a solid support,capturing the sample with PAPP-A in the sample.Anti-ProMBP antibody labeled with horseradish peroxidase was used as detecting antibody to test the the concentration of pregnancy-associated plasma protein A in human serum.Results The sensitivity of the assay we developed was 0.06 mIU/mL and it’s precision performed with mean CVs was 4.59%.The assay we developed was compared with enzyme immunoassay kit abroad,the result showing no significant difference between the two methods.Conclusion Using a particular paired double-antibody,the assay we established is sensitive and accurate.There is no significant difference between the assay and business kit abroad.Technical innovation is done in the paired antibody choosing and conjugate purification.更多还原
