长春瑞滨对人肺腺癌973细胞放射增敏作用的实验研究

【摘要】 目的:探讨长春瑞滨(NVB)对非小细胞肺癌细胞放射效应影响及其与放射的最佳结合序贯。方法:以指数生长期的人肺腺癌973细胞为研究对象,采用克隆形成分析法进行0.1nM及1nMNVB与放射不同结合序贯实验,研究NVB与放射结合效应,利用"多靶单击"数学模型(SF=1-(1-е-D/D0)N)进行曲线拟合,获得细胞存活曲线及平均致死剂量、准阈剂量、D0比和增敏比等参数,进行效应评估。结果:0.1nM及1nMNVB照前、照后给药组的增敏比(SER)分别为0.957和0.989,1.295和1.042;0.1nM及1nMNVB照前、照后给药组的D0比分别为1.073和1.099,1.374和1.106。表明0.1nMNVB照前、照后给药时均无放射增敏作用;1nMNVB照前、照后给药时均与放射效应具有较明显协同作用,且照前给药组的协同作用明显强于照后给药组。结论:0.1nMNVB不具有增强放射效应的作用,1nMNVB具有较明显放射效应增强作用,且照前给药组的协同作用明显强于照后给药组。

【Abstract】 Objective:To investigate whether navelbine(NVB)can enhance the effect of radiation to human non small cell lung cancers and the optimal combined schedule of NVB and radiation.Methods:Human lung cancer 973 cells on exponential growth were used for study.The effects of NVB combining radiation to 973 cells were tested by clonogenic assay,and the experiments including various schedules of 0.1nM NVB and 1nM NVB combining radiation,"multi target single hit survival model"(SF=1-(1-е-D/D0)N)was used for dealing with data,then the cell surviving curve,D0,Dq,SF2,D0 ratio and SER were obtained.Results:The SER of 0.1nM NVB and 1nM NVB administrated before and after irradiation respectively were 0.957 and 0.989,1.295 and 1.042;D0 ratio of 10nM GEM and 100nM GEM administrated before and after irradiation respectively were 1.073 and 1.099,1.374 and 1.106.0.1nM NVB administrated before and after irradiation had no radiosensitization.The combinations of 1nM NVB administrated before and after irradiation with radiation result in obvious synergism,and the synergism of 1nM NVB followed by irradiation was stronger than reversed treatment sequence.Conclusion:0.1nM NVB has no radiosensitization;1nM NVB can enhance the effect of radiation,and the enhancement of 1nM NVB followed by irradiation is stronger than reversed treatment sequence.
