固相萃取-反相HPLC荧光法快速测定血浆中氟罗沙星

【摘要】 目的建立固相萃取-反相HPLC荧光法快速测定人血浆中氟罗沙星的方法。方法采用C18固相萃取小柱萃取血浆中的氟罗沙星。色谱柱为ODS-BP柱(5μm,250 mm×4.6 mm),流动相为含1.31 g/L L-异亮氨酸和0.80 g/LCuSO4的水溶液-甲醇(80∶20,V∶V),激发波长为280 nm,发射波长为504 nm,流速为0.8 ml/min。结果该方法在0.4-20μg/ml浓度范围内线性关系良好(r=0.999 3),最小检测限50 ng/ml,萃取回收率和方法回收率分别为89.52%和100.45%,高、中、低3个浓度的日内RSD为3.2%,日间RSD为1.5%。结论该法血样处理简单,干扰小,灵敏度高,精密度好,可用于该药物体内监测的快速测定。

【Abstract】 Objective To set up a new method for quickly determining fleroxacin in human plasma by SPE and RP-HPLC with fluorescence detection.Methods Fleroxacin in human plasma was extracted by SPE with C18 SPE column,and then the quantities of fleroxacin were determined by RP-HPLC.The chromatography column was a ODS-BP column(5 μm,250 mm×4.6 mm).And the mobile phase was made up of the water solution including 1.31 g/L L-isoleucine and 0.80 g/L CuSO4 and methanol(80∶20 ,V∶V).The excitation and emission wavelengths were 280 nm and 504 nm.The flow rate was 0.8 ml/min.Results The linear range of the method was 0.4-20 μg/ml(r=0.999 3),the detection limit was 50 ng/ml(S∶N=3∶1), and the extraction recovery rate and the method recovery rate were 89.52% and 100.45%,respectively.The within-day RSD was 3.2%,and the between-day RSD was 1.5%.Conclusion The method is simple in the pretreatment of the bio-samples,sensitive and precise.And it can be applied to rapid determination of fleroxacin.
