多发性骨髓瘤p16基因甲基化与其预后的相关性
Relationship between Methylation of p16 Gene and Prognosis of Multiple Myeloma
【摘要】 目的探讨多发性骨髓瘤p16基因甲基化与其预后之间的相关性。方法采用甲基化敏感的限制性内切酶联合聚合酶链反应方法(REP法),检测52名多发性骨髓瘤(MM)患者骨髓细胞p16基因第1外显子的甲基化状态,并检测存在p16基因第1外显子甲基化的MM患者p16基因的mRNA转录水平。对这52名患者的临床资料及预后因素进行分析。结果52名MM患者中,p16基因启动子区第1外显子甲基化率为53.84%(28/52)。存在p16基因第1外显子甲基化的MM患者中,p16基因mRNA阴性率为75%。p16基因甲基化患者的预后不良因素明显高于非甲基化患者。结论多发性骨髓瘤p16基因甲基化与其预后有关。
【Abstract】 Objective To explore the relationship between methylation of p16 gene and the prognosis of multiple myeloma(MM).Methods Fifty-two patients with multiple myeloma were tested for methylation status of the first exon of p16 gene by restriction enzymeralated polymeras chain reaction(REP),and those with positive results were detemined for transcriptioin level of mRNA.The clinical data and prognostic factors of the 52 patients were analyzed.Results Twenty-eight(53.84%)cases of methylation of the first exon of p16 gene were observed,of which the negative rate of mRNA was 75%.The levels of factors adversely influencing the prognosis in the patients with methylation of the first exon of p/G gene were significantly higher than those in the patients without methylation.Conclusion The hypermethylation of p16 gene was related to the prognosis of multiple myeloma.
